Objective: To study the expression of mi R-152-3p and PTEN in Stanford-A acute aortic dissection and the effect of their interaction on the occurrence of acute aortic dissection.Method: 1)Aortic tissue samples were collected from 30 patients with aortic dissection surgery in our hospital as dissection group and 30 patients with coronary artery bypass graft surgery as control group.The relative expression of mir-152-3p was detected by qRT-PCR.Western blot to detect the relative expression of PTEN protein.In the histopathological section,HE staining,Masson staining and Immunohistochemical staining were used to observe the morphology of aortic wall.2)Human aortic smooth muscle cells were cultured and transfected into mir-152-3 pmic,mir-152-3 pmic control,mir-152-3 pibitor control,mir-152-3 pibitor control,blank control group(respective mimic group,mimi-con group,inhibitor group,inhibitor-con group,nc group).draw the level curve of mir-152-3p expression,select the best time point to collect the cells.The expression of pten protein and related protein in pten/akt pathway was detected by western blot,the apoptosis of the cells after transfection was detected by flow cytometry.Result: 1)The expression of mir-152-3p in dissection group was significantly lower than that in control group(p <0.05).Westernblot results showed that pten was expressed in the dissection group and the control group,and the expression of pten in dissection group was significantly higher than that in control group(p <0.05).The results of HE staining and Masson staining showed that the arrangement and morphology of media of aortic dissection in the dissection group were obviously disordered,multiple elastic fiber breakage and myxoid degeneration,the collagen fibers proliferated obviously,The structure of media in the control group was compact and the shape was regular,there was no obvious fiber fracture and myxoid degeneration and proliferation of elastic fibers.2)Cell experiments showed that overexpression or inhibition of miR-152-3p expression could affect the expression of PTEN at mRNA and protein level,in the mimic group overexpressing mir-152-3p,the expression of pten and pro-apoptotic gene bax was significantly down-regulated compared with that of nc group and mimi-con group,while the expression of pro-apoptotic gene bcl2 was significantly up-regulated;In inhibitorgroup,the expression of PTEN and pro-apoptotic gene BAX was significantly up-regulated compared with NC group and inhibitor-con group,while the expression of anti-apoptotic gene BCL2 was significantly down-regulated.Moreover,the results of flow cytometry showed that transfection of miR-152-3p mimic inhibited apoptosis,while transfection of mi R-152-3p inhibitor promoted apoptosis.Conclusion: MiR-152-3p should promote the apoptosis of smooth muscle cells by regulating the expression of PTEN,and then promote the occurrence of aortic dissection.
