Benign prostatic hyperplasia(BPH)is a common chronic disease with high incidence of a disease,which seriously affect the patients' health and life quality.Pumpkin seed oil is a unique flavor oil with abundant nutrition and effective components,which has functions for suppression of benign prostatic hyperplasia.However,the active ingredient and resistance mechanism of anti-BPH is not clear.This study analyzed the components of the pumpkin seeds oil and investigated the extraction process of phytosterol from pumpkin seed oil(PPS).The effects and potential mechanisms of PPS were studied through treating on BPH rats induced by testosterone propionate(TP).1.The bioactive components were analyzed by GC-MS and HPLC.The unsaponifiables extract process was determined.The results shown that the pumpkin seed oil mainly contained 4 kinds of unsaturated fatty acid with a total content of 74.73%,including the cismonounsaturated fatty acid(29.14%)and polyunsaturated fatty acid(45.54%).Pumpkin seeds oil was rich in phytosterol and squalene,and the content of squalene was 5 490 ?g/g.Extraction and preliminary purification of PPS were conducted by saponification-solvent crystallization.Phytosterol content is 94.8% by UV detection.2.Five fractions including three compounds were seperation by preparative HPLC,and each fractions were authenticated by GC-MS and NMR.?7-phytosterols are the main constituent in PPS: 24?-ethylcholesta-7,22,25-trienol,24?-ethylcholesta-7,25(27)-dien-3-ol(?7-avenasterol),(3?,5?,24Z)-Stigmasta-7,24(28)-dien-3-ol,?7-Campeterol,?7-sitosterol.And two ?5-phytosterols includes ?5-Campeterol,?5-sitosterol.3.The SD rats induced by exogenous hormone TP were set as BPH model.PPS gained by saponification-solvent crystallization from pumpkin seeds oil was considered as raw material and was used to treat on TP-induced BPH rats.Results shown:(1)PPS treatment significantly decreased prostate tissue weight and prostate index(p<0.05).(2)The size of prostate tissue reduced and the hyperplastic nodule recovered after treated by PPS.After treated by PPS,the epithelium and lumen were improved.(3)The PPS significant reduced the mRNA and protein expression of 5AR and ARs.(4)The PPS had the capability to reduce the mRNA expression of IGF,EGF,bFGF and to enhance the mRNA expression of pro-apoptotic factor TGF-?1,Bax/ Bcl-2.(4)PPS significantly reduced the level of p-ERK and increased the level of p-JNK.PPS suppressed BPH by preventing cell proliferation and promoting cell apoptosis.
