| The edible fungi have attracted much attention in the field of health food because of their pharmacological activities,such as enhancing immunity,anti-tumor,anti-oxidation,anti-inflammation,lowering blood sugar and be good at stomach and liver.Moreover,its natural medicinal active ingredients have the advantages of effect-enhancing and toxicity-reducing,good safety and fewer side effects,and have great application value in the field of health food and biomedicine.Sarcodon aspratums,cordyceps militaris and truffles are edible fungi,which are rich in nutrients and great benefit to human health.In order to facilitate nutrition intake and disease prevention,this paper intends to use water extraction method to extract sarcodon aspratums,cordyceps militaris and truffles,and then make the extract into tablets.This article main research content is as follows:1.Establishment of methodology for polysaccharide content determination in the extract.First of all,by using the water extraction method,extract the Sarcodon aspratums,cordyceps militaris and truffles.Then,according to the requirements for the determination of polysaccharide content in Chinese Pharmacopoeia(2015),the vitriolic acid-phenol method was selected to measure the polysaccharide content in the extraction of Sarcodon aspratums,cordyceps militaris and truffles.The results show that the extraction has the maximum UV absorption peak at 490 nm,and the sample average recovery is 104.54%.The experimental results show that the vitriolic acid-phenol method is simple,accurate,stable and reproducible,and can be used as a method for the determination of polysaccharides from the extraction of Sarcodon aspratums,cordyceps militaris and truffles.2.Establish and optimize the extraction method of Sarcodon aspratums,cordyceps militaris and truffles.The filtrate the extraction method by single factor experiment was optimized.The extraction temperature,extraction time,ratio of material to liquid and extraction times were discussed.The results of single factor experiment show that the polysaccharide content in the extraction is the highest when the extraction temperature is 90℃,the ratio of material to liquid is 1: 50,and the times of extraction is 7.Under these conditions,the extraction method is the best.Then using the orthogonal test optimize the extraction process,take the temperature of extraction,the ratio of material to liquid and extraction times as the evaluation criterion.The results of orthogonal test show that the extraction times has a greatest influence on the polysaccharide content in the extraction,followed by the ratio of material to liquid,and the least influence on the temperature of extraction.Therefore,the optimal extraction method is to mix the mixed bacterial powder and deionized water in a ratio of 1:50,conduct ultrasonic treatment for 15 min,and take a water bath at 90℃ for 1 h for 7 times.3.Qualitative analysis on the compound water extracts from Sarcodon aspratums,cordyceps militaris and truffles by UPLC-Q-Exactive HF-MSUsing UPLC-Q-Exactive HF-MS,analyze the samples comprehensively.The experimental results show that there are 68 compounds in the water extract of Sarcodon aspratums,cordyceps militaris and truffles,including a variety of carboxylic acid derivatives and organonitrogen compound,of which the organic peroxide concentration is the highest,followed by benzene and its substituted derivatives.4.The tablet preparation and quality detection of the extraction of Sarcodon aspratums,cordyceps militaris and truffles.Prepare tablets and conduct the quality detection.According to the tests,the tablet is complete,smooth,uniform in color,and no hemp spots.The weight difference is <5.0%,and tablets can disintegrate in 15 minutes,the hardness is moderate,and the weight loss is <1%,with no fracture,crack or comminution,which lay a theoretical foundation for the further production and application of the tablet preparation of Sarcodon aspratumss,cordyceps militaris and truffles extracts. |
