AIM:To investigate the possible mechanism of platinum resistance in non-small cell lung cancer by comparing the expression of Sall4 in non-small cell lung cancer and platinum-resistant non-small cell lung cancer.Methods:The control group was A549 cell line,and the experimental group was cisplatin-resistant A549/DDP cell line.The cells were cultured with F12-K containing 10%fetal bovine serum and main-tained in a humidified atmosphere of 5%CO2 in air at 37°C.The expression of Sall4 protein was observed by cellular immunofluorescence.The expression of Sall4 mRNA was detected by reverse transcription-polymerase chain reaction?RT-PCR?.Results:The results of cellular immunofluorescence showed that Sall4 protein mainly expressed in the cytoplasm of non-small cell lung cancer cells.The average fluorescence intensity of the experimental group?23.87±1.04?was significantly higher than that of the control group?17.63±3.36?,and the differences between the two groups were significantly different?P<0.05?,RT-PCR results showed that the gray values of Sall4 mRNA in the experimental group and the control group were 1.06±0.15 and 0.65±0.13,respectively.The comparison between the two groups was statistically significant?P<0.05?.Conclusion:Sall4 may be involved in the acquired platinum resistance of non-small cell lung cancer.
