Objective To investigate the effects of sevoflurane postconditioning on oxidative stress and mitochondrial function of hippocampus in a rat model of hemorrhagic shock and resuscitation.Methods Male SD rats were randomly divided into Sham surgery group?Sham group?,shock and resuscitation group?Shock group?,and 2.4%sevoflurane postconditioning group?Sevo group?.Hemorrhagic shock and resuscitation animal models were prepared by withdrawing 40%of the total blood volume of the rat within30 min through the right carotid artery.After observation for 1 h,the blood was received through the left jugular vein in 30 min.The Sevo group inhaled 2.4%sevoflurane immediately after reperfusion,and the Sham group and Shock group inhaled 95%O2-5%CO2 mixture at the corresponding time period respectively.The mean arterial pressure?MAP?was recorded at an interval of 10 min,and the arterial blood was collected for blood gas analysis at the time of the beginning of bleeding?Baseline,T0?,the finish of blood withdrawing?30 min after the catheterization in Sham group,T1?,the start of blood reperfusion?90 min after the catheterization in Sham group,T2?,and the end of blood reperfusion?120 min after the catheterization in Sham group,T3?.Blood gas analysis includes buffuer excess?BE?and p H and lactic acid concentration?Lac?.After 24h of operation,the content of malonaldehyde?MDA?and reactive oxygen species?ROS?in hippocampus was determine.The activity of superoxide dismutase?SOD?and mitochondrial respiratory chain complex I/II/III/IV in hippocampal mitochondria were detected,as well as the activity of mitochondrial permeability transition pore?m PTP?.The level of mitochondrial membrane potential?MMP?of hippocampus was tested by a kind of assay kit.Western blot was used to determine the relative expression of silencing information regulatory factor 1?SIRT1?and peroxisome proliferator-activated receptor?coactivator-1??PGC-1??in hippocampal tissues.Results Compared with Sham group,as for Shock group,the content of MDA and ROS increased,the level of MMP and the activity of SOD and m PTP and mitochondrial respiratory chain complex I/II/III/IV decreased,and the relative expression of SIRT1 and PGC-1?increased?P<0.05?.Compared with Shock group,the content of MDA and ROS dereased,the level of MMP and the activity of the SOD and m PTP and mitochondrial respiratory chain complex I/II/III/IV increased,and the relative expression of SIRT1 and PGC-1?increased increased in Sevo group?P<0.05?.Conclusion Sevoflurane postconditioning can reduce oxidative stress and mitochondrial dysfunction of hippocampus in a rat model of hemorrhagic shock and resuscitation,the mechanism of which may be related to the up-regulation of SIRT1and PGC-1?expression...
