Objective: To establish a severe acute pancreatitis(SAP)rat model and study the effect of olive oil lipid emulsions(OOLE)on the intestinal mucosal barrier of SAP rats,and provide theoretical support for clinical parenteral nutrition therapy.Methods: All rats underwent unilateral external jugular venous catheterization.After 48 hours,Thirty male SD rats were randomly divided into 3 groups: sham operation group(Control group),model group of severe acute pancreatitis(SAP group)and olive oil lipid emulsions intervention group(OOLE group).Rats in the control group and SAP group were pumped with normal saline daily,and the OOLE group was pumped with OOLE daily.Five days later,rats in the control group underwent sham abdominal surgery,and the SAP group and the OOLE group underwent retrograde pancreaticobiliary duct injection with sodium taurocholate for modeling surgery.After 24 hours,the abdominal cavity of the rats was observed,meanwhile,ascites,blood of aortaventralis,specimens of small intestine and pancreas were collected.The levels of blood amylase and lipase were tested by automatic biochemical facility.The levels of tumor necrosis factor-?(TNF-?),interleukin-6(IL-6)and diamine oxidase(DAO)in serum were tested by enzyme-linked immunoassay(ELISA).The pathological changes of pancreas,small intestine tissues and the villus crypt units of small intestine were measured respectively by HE staining.The intestinal epithelial proliferating cell nuclear antigen(PCNA),apoptosis-related protease Caspase-3 and cell tight junction protein ZO-1 was analyzed and quantified by Image J and Image-Pro plus 6.0 software.Results:(1)Compared with Control group,obvious intraperitoneal inflammation of rats in SAP group had been infected after modeling,and the amount of ascites was significantly increased(P<0.01).The levels of blood amylase,lipase,serum IL-6 and TNF-? were significantly increased(P<0.01);HE staining showed large-scale necrosis of pancreatic tissue and infiltration of inflammatory cells,and the value ofintestinal villi height and crypt depth were significantly decreased(P<0.01).(2)Compared with the SAP group,the blood amylase,lipase and serum IL-6 levels in the OOLE group were not statistically different(P>0.05).TNF-? level was decreased(P<0.05),intestinal mucosal epithelial cells proliferation rate(PCNA positive rate)was increased,apoptosis rate(Caspase-3 positive rate)was decreased(P<0.05),and intestinal villi height and crypt depth values were increased(P<0.05).Intestinal mucosa tight junction protein ZO-1 level was increased(P<0.05),DAO level was decreased(P <0.05),both led to intestinal permeability decreased.Conclusions:(1)Inflammation-related indicators of SAP model rats were increased,meanwhile,damage to the intestinal mucosal barrier by intestinal epithelial cells and tight intercellular connections was found.(2)OOLE can promote the proliferation and reduce apoptosis of intestinal mucosal epithelial cells,reduce the damage of intestinal villous crypt units,increase intestinal mucosal tight junction protein expression to maintain intestinal permeability,thereby playing a protective role on the intestinal mucosal barrier of SAP rats.
