Objective To understand the effect of intermittent hypoxia on the expression of neural factors in rat brain and its possible mechanism,and to explore the protective effect of Sofren injection on rat brain.Methods 45 healthy adult male SD rats were randomly divided into 5 groups:normoxia control group(NC),intermittent hypoxic group(IH),intermittent hypoxic+low-dose sofren injection group(IH+low-dose SI),intermittent hypoxic+medium dose sofren injection group(IH+medium-dose SI),intermittent hypoxic+high-dose sofren injection group(IH+high-dose SI),9 rats in each group.The rats were placed in the intermittent hypoxia chamber.The chamber of the normoxia control group was filled with compressed air,and the chamber of the intermittent hypoxia group was intermittently filled with compressed air,nitrogen and oxygen.Each sofren injection intervention group rats was intraperitoneally injected with the corresponding dose of sofren injection 1 hour before entering the chamber,and the other groups were injected with the same amount of normal saline.The rats were put to death after 45 days,nitric oxide(NO),superoxide dismutase(SOD)and malondialdehyde(MDA)were deceted by colorimetry,erythropoietin(EPO)and hypoxia-inducible factor(HIF-1?)were detected by enzyme-linked immunosorbent assay,brain derived neurotrophic factor(BDNF)and nerve growth factor(NGF)were detected by polymerase chain reaction.The correlation between oxidative stress,hypoxia and neural factors was analyzed.Results Compared with the NC group,the concentration of NO and SOD in brain tissue of rats in the HI group decreased(P<0.01),the concentration of MDA,EPO,HIF-1? increased(P<0.01),and the expression of BDNF and NGF increased(P<0.01).Compared with the IH group,the contents of NO and SOD in the rabraintissues of the IH + SI increased(P < 0.05),the contents of MDA,EPO and HIF-1?decreased(P < 0.05),and the expressions of BDNF and NGF genes decreased(P <0.01).IH+high-dose SI showed the most significant improvement in a dose-dependent manner.Correlation analysis showed that the NGF was negatively related to NO(r=-0.333 P=0.047)and SOD(r=-0.805 P=0.000).and positively correlation with MDA(r=0.565 P=0.000),EPO(r=0.711 P=0.000),HIF-1?(r=0.786 P=0.000),BDNF expression level was negatively correlated with NO(r=-0.516 P=0.001),SOD(r=-0.861 P=0.000),and positively correlated MDA(r=0.490 P=0.002),EPO(r=0.702 P=0.000),HIF-1?(r=0.697 P= 0.000).Conclusion Intermittent hypoxia could significantly reduce the concentration of NO and SOD in rat brain tissue,and the concentrations of MAD,HIF-1?and EPO were significantly increased.Correlation analysis suggested that NGF was negatively correlated with NO and SOD,and positively correlated with MDA,EPO and HIF-1?.The expression level of BDNF was negatively correlated with NO and SOD,and positively correlated with MDA,EPO and HIF-1?.sofren injection can alleviate the oxidative stress injury induced by intermittent hypoxia,alleviate the chronic hypoxia injury,and reduce the activity of neural factors in the brain by inhibiting the production of prooxide and increasing the level of antioxidant,so as to prevent nerve injury,promote nerve repair and reduce the damage effect of intermittent hypoxia on brain cells.
