Background: A large amount of literature shows that circular RNA can participate in the regulation of gastric cancer development.This study aims to detect the expression of hsacirc0010882 in the serum and cell lines of gastric cancer patients,and to observe the expression of this molecule on gastric cancer cell proliferation and metastasis,and the effects of behaviors are explored for the possible role of hsacirc0010882 in the regulation mechanism of gastric cancer;Clarify the impact of hsacirc0010882 on gastric cancer cells,verify their possible regulatory mechanism,and provide new ideas for the treatment and management of gastric cancer patients.Methods: 1.The expression of hsacirc0010882 in gastric normal mucosa epithelial cells and gastric cancer cells were detected by real-time quantitative PCR,and the expression difference between the serum of gastric cancer patients and normal human serum was analyzed.The correlation between the molecule and clinicopathological characteristics was analyzed.2.Establish hsacirc0010882 over-expressing and under-expressing gastric cancer cell models using lentivirus infection technology.Gain-of-function and loss-of-function assays were used to investigate the physiological function of hsacirc0010882 in GC cells in vitro in the context of proliferation,apoptosis,migration,and invasion.3.Combined with the results of in vitro experiments and the literature,find a signal pathway that hsacirc0010882 may participate in regulation,and verify whether hsacirc0010882 can regulate gastric cancer cells through this pathway using western blot.Results: 1.The expression level of hsacirc0010882 in all four gastric cancer cell lines was higher than that of normal gastric epithelial cells.Among them,hsacirc0010882 expressed the highest in MKN-45,SGC-7901 cell lines.The expression of hsacirc0010882 in the serum of gastric cancer patients is higher than that in normal human serum,and it is positively correlated with the clinical stage of gastric cancer patients,and the tumor size is positively correlated;Hsacirc0010882 high-expressing gastric cancer patients have a lower survival time than hsacirc0010882-low gastric cancer patients.2.The results of in vitro experiments show that when hsacirc0010882 is overexpressed in gastric cancer cells,it can significantly promote cell proliferation,migration,and invasion ability,and has no significant effect on the apoptosis rate;When hsacirc0010882 is low expressed in gastric cancer cells,it can inhibit cells proliferation,migration,invasion ability,and promote apoptosis;3.Based on the results of in vitro experiments,a large number of related literature were consulted.Finally,PI3K/Akt/mTOR was used as a signal pathway that hsacirc0010882 may participate in regulation.Western blot was used to verify the activation of the PI3K/Akt/mTOR signaling pathway.The results showed that hsacirc0010882 overexpression could promote the expression of the PI3K/Akt/mTOR signaling pathway.Conclusion: hsacirc0010882 is highly expressed in gastric cancer,and the expression level is positively correlated with clinical stage and tumor size,which can be an independent predictor of prognosis in patients with gastric cancer.hsacirc0010882 positively regulates malignant biological behaviors such as proliferation,invasion,and migration of gastric cancer cells.hsacirc0010882 promotes the progression of gastric cancer by activating the PI3K/Akt/mTOR signaling pathway.
