Objective:Atherosclerosis is a diffuse inflammatory vascular disease related to age,sex,diet,lifestyle,blood pressure,blood glucose,blood lipid and other factors,it's an arterial vascular disease which can affect localized or diffuse arteries of the whole body.Atherosclerosis can occurs in coronary arteries,intracranial arteries,renal arteriesother important blood vessels,and often causes serious consequences.Therefore,the basic and clinical study of atherosclerosis has always been the foucs of attention.Among them,inflammatory response and proliferation of smooth muscle cells are the important pathological basis of atherosclerosis.Luteolin has received increasing due to its therapeutic effect on a variety of tumors,especially its role in cell proliferation and migration,as well as its relationship with inflammatory response.This subject aims to observed the effect of luteolin on the physiological fuction of human aortic vascular smooth muscle cells,and study the endoplasmic reticulum stress activation is one of the possible regulatory mechanism,so as to explore mignonette element by endoplasmic reticulum stress influence theeffect of smooth muscle cells migration,to seek a new theoretical basis for blood vessel damage repair,for the Chinese medicine treatment of atherosclerosis provide a new clinical idea.Methods:HASMCs was selected as the research object,and treated with luteolin at concentrarions of 0,6.25,12.5,25and 50umol/L respectively to dectect the migration and endoplasmic reticulum stress levels of HASMCs.The endoplasmic reticulum stress inhibitor 4-PBA was treated at different concentrations and at different times.The difference between HASMCs migration and endoplasmic reticulum stress levels after treatment with luteolin with 4-PBA and without 4-PBA was compared.Cell scratch was used to detect the migration of HASMCs.RT-PCR was used to detect the mRNA expression and Westren blotting was to detect the protein rxpression.Results:(1)Luteolin can inhibit the migration of HASMCs in a concentration dependent manner.(2)Luteolin promotes endoplasmic reticulum stress in HASMCs.(3)Endoplasmic reticulum stress inhibitor 4-PBA can enhance the migration ability of HASMCs.(4)Inhibiting endoplasmic reticulum stress can weaken the inhibitory effect of luteolin on the migration of HASMCs.Conclusion:(1)Luteolin negatively regulates the migration of HASMCs.(2)Luteolin positively regulates the endoplasmic reticulum stress of HASMCs.(3)Inhibiting endoplasmic reticulum stress can weaken the inhibitory effect of luteolin on the migration of HASMCs.
