Objective:This study was conducted to analyze the relationship between peroxiredoxin 3 and blood glucose and insulin in patients with gestational diabetes mellitus(GDM),which would be helpful to understand the role of PRX3 in the development and progression of GDM.Method:(1)Pregnant women at 24-28 gestational weeks underwent oral glucose tolerance test(OGTT).Patients who meet the diagnostic criteria for GDM were enrolled as test subjects,and age-matched pregnant women that did not reach the OGTT threshold were registered as the control subjects.After routine examination of blood glucose and insulin,serum PRX3 was detected by enzyme-linked immuno sorbent assay.All pregnant women enrolled in the study were admitted to hospital at 40 weeks and their fasting blood was obtained to measure blood glucose,insulin,and serum PRX3 levels.(2)Western blotting was used to detect the oxidation of PRX3 in peripheral blood of three time points of OGTT.(3)Immunohistochemistry was applied to compare the expression of PRX3 in placentas of GDM patients and controls.Result:(1)Thirty GDM patients and thirty control subjects were included in the present study.The blood glucose at fasting,1h,and 2h of OGTT was significantly higher in GDM patients than in controls(P<0.001,P<0.05,and P<0.05 respectively).The serum insulin at 1h of OGTT was lower in GDM patients than in controls(P<0.05),while no difference was observed at fasting and 2h of OGTT.There was no significant difference in serum PRX3 between the two groups at three time points of OGTT.(2)The serum PRX3 of GDM patients was significantly increased at 2h of OGTT as compared to fasting level(P<0.05),and the serum PRX3 at 2h was positively correlated with the glucose level at 1h of OGTT(r=0.760,P=0.011).(3)Serum PRX3 and FPI are positively correlated with Gestational week during pregnancy,statistically significant(r>0,P<0.05)?(4)The levels of oxidized PRX3 in peripheral blood cells,together with oxidized PRX1 and PRX2,were significantly increased in GDM patients as compared to controls at 1h and 2h of OGTT(P<0.05).(5)There was no significant difference in placental staining index of PRX3 between GDM patients and control subjects(F=2.360,P>0.05).Conclusion:Our results indicate the limitation of insulin secretion in GDM patients,leading to high blood glucose and subsequent oxidative stress.PRX3 plays an active role in the response to the oxidative stress,which would attenuate the progression of GDM.Since a substantial number of GDM patients did not control blood glucose very well and some control subjects showed high blood glucose at term pregnancy,it might be interpretable that no significant difference was found in placental staining index of PRX3 between GDM patients and control subjects.
