Objective:The different expression of long non-coding RNA CASC2 in esophageal squamous cell carcinoma tissues and their adjacent tissues were detected,to explore its pathogenesis in esophageal squamous cell carcinoma.Methods:Firstly,PCR was used to detect the expression levels of lncRNA CASC2 in esophageal squamous cell carcinoma and para-carcinoma tissues.More than that,the expression of lncRNA CASC2 in a variety of esophageal squamous cell lines and normal epithelial cell lines was detected by qPCR.Secondly,based on the above research,in order to explore the lncRNA CASC2 its biological functions in esophageal cancer,we respectively in the esophageal squamous cancer cells EC 109 transfection CASC2 siRNA after training,and collect the EC 109 cells,by qPCR experiment testing cells IncRNA CASC2 expression and transfection efficiency,observe the lncRNA CASC2 effect on esophageal cancer cell proliferation at a cellular level.Results:The experimental results of this study showed that the expression of lncRNA CASC2 in esophageal squamous cell carcinoma was decreased compared with that in esophageal squamous cell carcinoma.At the same time,through the detection of lncRNA CASC2 in a variety of esophageal squamous cell lines and normal epithelial cell lines,it was confirmed that the expression of lncRNA CASC2 in esophageal squamous cell lines was also lower than that in normal esophageal epithelial cells.However,the overexpression of CASC2 in esophageal squamous cell carcinoma can reduce cell proliferation,and the decreased expression can promote the proliferation of cancer cells,indicating that lncRNA CASC2 plays a role as an anti-tumor factor in esophageal squamous cell carcinoma and inhibits the proliferation of esophageal squamous cell carcinoma to some extent.In addition,in order to explore the effect of CASC2 on esophageal squamous cell carcinoma in animal culture,we constructed subcutaneous tumor mice of esophageal squamous cell carcinoma,and found that the in vivo experimental results were consistent with the cell results,that is,increased CASC2 expression could slow down cell proliferation and inhibit tumor formation,and decreased CASC2 expression could accelerate cell proliferation and promote tumor formation.The effects of CASC2 expression on apoptosis in esophageal cancer tumor tissues were assayed.Conclusion:The down-regulated expression of lncRNA CASC2 in both esophageal squamous cell carcinoma tissues and cells has preliminarily confirmed that CASC2 plays a role as an anti-tumor factor in esophageal squamous cell carcinoma,in the meantime its increased expression can inhibit the proliferation of esophageal squamous cell carcinoma tumor cells,thus inhibiting the formation of esophageal tumor.
