| Objective:By examining the expression of BTG2 and SEMA3D in laryngeal cancer tissue and normal tissue adjacent to carcinoma,combined with clinical data,we will analyzed the relationship of its expressions and clinicopathological characteristics and prognosis.Methods:1.Seventy-six laryngeal cancer tissues and normal tissues adjacent to carcinoma in Otolaryngology-Head and Neck Surgery of Affiliated Hospital of North Sichuan Medical College were obtained from January 2015 to August 2018.Immunohistochemistry staining was used to detect the expression of BTG2 and SEMA3D.The relationships of its expression with clinicopathological characteristics and prognosis were analyzed.SPSS22.0 was used for statistical analysis,and the Chi-square test,the continuous correction Chi-square test and Fisher Precision test were used to analyze the positive rate of proteins between different clinicopathological characteristics.2.The prognosis of 67 cases of laryngeal cancer from January 2015 to August 2018 was obtained through review,internet and telephone follow-up.Log-Rank test and Cox regression analysis were used to analyze the correlation between the proteins and the prognosis of patients,and the survival curve was drawn by Kaplan-Meier method.The statistical test level was taken as α=0.05,P<0.05 was considered statistically significant.Results:1.Immunohistochemistry showed that the positive expression of the BTG2 and SEMA3D was diffuse tan,and the BTG2 was located in the cytoplasm,and the SEMA3D was located in the cytoplasm and part of membrane.The respectively positive expression of BTG2 in laryngeal cancer tissues and normal tissues adjacent to carcinoma was 38.16%and 90.79%,and the difference was statistically significant(P<0.05).The respectively positive expression of SEMA3D in laryngeal cancer tissues and corresponding adjacent tissues was 26.32%and 72.37%,and the difference was statistically significant(P<0.05).2.The respectively positive rates of BTG2 expression in age less or equal 50 and over were 18.18%and 46.30%,P<0.05 the difference was statistically significant.The respectively positive rates of BTG2 in high differentiation,medium differentiation and low differentiation were 48.00%,19.05%and 20.00%,and the difference was statistically significant(P<0.05).The respectively positive rates of BTG2 in patients with and without cervical lymph node metastasis were 24.24%and 48.84%,and the difference was statistically significant(P<0.05).3.The respectively positive rates of SEMA3D in TNM stage Ⅰ-Ⅱ and Ⅲ-Ⅳ were 39.39%and 16.28%,and the difference was statistically significant(P<0.05).The respectively positive rates of SEMA3D in patients with and without cervical lymph node metastasis were 12.12%and 37.21%,and the difference was statistically significant(P<0.05).4.There was no significant difference between the expression level of BTG2 and gender,clinical tumor location and TNM stage(P>0.05).There was no significant difference between the expression level of SEMA3D and age,gender,clinical tumor location and pathological grade(P>0.05).5.Univariate Log-Rank test found that TNM stage,cervical lymph node metastasis,BTG2 expression,and SEMA3D expression were associated with patients’ overall survival(P<0.05).Multivariate Cox regression analysis revealed cervical lymph node metastasis and the expression of BTG2 and SEMA3D correlated with the overall survival of the patients(P<0.05).Conclusion:1.The positive rates of BTG2 and SEMA3D in laryngeal cancer tissue were lower than normal tissue adjacent to carcinoma.2.The expression of BTG2 was closely related to age,pathological grade and cervical lymph node metastasis of laryngeal cancer.The increasing of age,the higher of pathological grade and no cervical lymph node metastasis,the higher the positive expression of BTG2 in patients.3.The expression of SEMA3D was closely related to TNM stage and cervical lymph node metastasis of laryngeal cancer.The early of TNM stage and no cervical lymph node metastasis,the higher the positive expression of SEMA3D in patients.4.Cervical lymph node metastasis and the expression of BTG2 and SEMA3D were associated with the overall survival of patients.The overall survival of the BTG2-positive group and the SEMA3D-positive group were significantly higher than the BTG2-negative group and the SEMA3D-negative group. |
