BackgroundThe treatment of osteosarcoma,a common malignant bone tumor,requires the complete removal of tumor tissue and the regeneration of resection-induced bone defects.Therefore,the implantation of multifunctional scaffolds with the capabilities of both anti-cancer therapy and bone regeneration is highly desirable.Materials and MethodsIn this study,hierarchically porous and mechanically strong ?-tricalcium phosphate/poly(lactic-co-glycolic acid)(TCP/PLGA)scaffolds with combined photothermal/chemo therapies and improved bone regeneration ability were produced by incorporating 2D black phosphorous(BP)nanosheets,doxorubicin(DOX)and bone morphogenetic protein-2 peptide(P24)in situ into TCP/PLGA scaffolds through novel cryogenic 3D printing.ResultsOn the one hand,the multifunctional scaffolds enabled both in vitro and in vivo osteosarcoma ablation through the synergistic effects of BP-related photothermal therapy and DOX-related chemotherapy.After 10 minutes of nir irradiation,the survival rate of MG63 cells cultured on BDPTP and BPTP scaffolds decreased to nearly 0%.On the other hand,in vitro osteogenic differentiation of rat bone marrow derived mesenchymal stem cells and in vivo regeneration of rat cranial bone tissues were achieved by the multifunctional scaffolds,owing to the synergistic effects of sustained release of BMP-2-like polypeptide and the existence of TCP in scaffolds.BMD values of the PTP,BPTP and BDPTP groups were significantly higher than those of the blank control group(423±60,400±50 and 385±50 vs.57.5±29,143±41 and 270±40 mg/mm3),(p<0.05).ConclusionsThis investigation provided a facile "one-step" method to fabricate customized scaffolds with both anti-cancer therapy and bone regeneration capabilities for treating osteosarcoma.
