The Expression And Clinical Significance Of LRP4 In Colorectal Cancer

Objective:Low-density lipoprotein receptor-related protein 4(LRP4)functions in the progression of many diseases including tumors.We aims to research the expression and role of LRP4 in colorectal cancer(CRC).Methods:We collected specimens of 45 CRC patients from January 2018 to September 2018.We collected CRC tissues and adjacent normal tissues.Immunohistochemistry(IHC),Western Blot and RT-qPCR were adopted to determine the LRP4 expression level in specimens.CRC cell lines LoVo and HCT-116 which had high LRP4 expression were selected for next experiments.LRP4 expression was knocked down by siRNA technology.Cell proliferation and migration tests were put into use to throw light on the function of LRP4 in CRC.Results:IHC results showed that LRP4 expressed high in 29(64.4%)CRC tissues and low in 16(35.6%)CRC tissues.LRP4 showed low expression in all normal tissues.The high expression level of LRP4 was in close touch with TNM staging(P=0.002),meanwhile having a bear on lymph nodes metastasis(P=0.005).RT-qPCR showed that LRP4 mRNA in CRC tissues turned out to have higher relative expression levels than that in normal tissues(0.765±0.185 vs.0.193±0.088,P<0.001).The results of cell proliferation test showed that after LRP4 knockdown and 4 days of culturing LoVo and HCT-116 cell lines,the proliferation capacity of the Si-LRP4 groups were both inferior to the Si-NC groups(96h absorbance values:LoVo:Si-LRP4 0.831±0.001 vs.Si-NC 1.236±0.003,P<0.001;HCT-116:Si-LRP4 0.832±0.001 vs.Si-NC 1.234±0.001,P<0.001).The results of cell migration test showed that after LRP4 knockdown and 24h of culturing LoVo and HCT-116 cell lines,the migration ability of Si-LRP4 groups were both significantly lower than that of the Si-NC groups.The differences of cell numbers in each visual field were statistically significant(LoVo:Si-NC 258.40±13.81 vs.Si-LRP4 111.20±10.78,P<0.001;HCT-116:Si-NC 324.20±19.66 vs.Si-LRP4 122.20±11.08,P<0.001).Conclusions:Compared to normal tissues,LRP4 takes on a higher expression level in CRC tissues.High expression level of LRP4 owns a relationship with TNM stages and lymphatic metastasis.LRP4 can expedite the proliferation and migration of CRC cells.
