Objectives:1.To explore the expression difference of CIP2A in glioma tissue and non-glioma brain tissue,further,analyze the relationship between the expression level of CIP2A in glioma tissue and clinical pathological grade,and to study the clinical significance of CIP2A in glioma.2.To investigate the effect of inhibition of CIP2A expression on the proliferation,invasion and migration of glioma cell lines,and to detect and analyze the effect of inhibition of CIP2A expression on epithelial-mesenchymal transition(EMT).Methods:1.Immunohistochemistry was used to detect the expression of CIP2A in glioma tissues and to analyze its correlation with clinicopathology.Differences in CIP2A expression between non-glioma tissues and glioma tissues were detected by database analysis and Western blot.2.Western blot and real-time quantitative PCR were used to detect the expression of CIP2A in glioma cell lines at the protein level and mRNA level,respectively,and two cell lines with relatively high CIP2 A expression were screened out.3.After inhibiting CIP2A expression in T98G and U251 by plasmid transfection,the expressions of EMT-related proteins(E-cadherin,N-cadherin,Vimentin,Snail)and MMP2、MMP9 were detected by Western blot.4.Application of immunofluorescence technology,CCK8,transwell and wound healing experiments to explore the effects of inhibition of CIP2A expression on cell proliferation,invasion and migration.Results:1.Immunohistochemistry and Western blot analysis showed that the expression of CIP2A in gliomas was higher than that in non-glioma tissues,consistent with the results of database analysis,and the expression was higher in high-grade gliomas(grades Ⅲ-Ⅳ)than in low-grade ones(Grade Ⅰ-Ⅱ).2.In the cell lines T98G,U251,the expression of N-cadherin,Vimentin,and Snail was the same as that of CIP2A after inhibiting the expression of CIP2A,and the expression was down-regulated,while the expression of E-cadherin was up-regulated.3.After suppressing the expression of CIP2A,the CCK-8 results showed that the proliferation ability of the inhibition group was lower than that of the control group;the immunofluorescence results showed that the edu proliferation markers of the inhibition group were significantly less than the control4.After inhibiting the expression of CIP2A in the transwell experiment,the number of T98G,U251 cells passing through the chamber to the serum side decreased,and the area of scratch healing was reduced in the scratch healing experiment.Conclusions:1.CIP2A is highly expressed in gliomas,and its expression level may be related to survival prognosis.2.In vitro experiments confirmed that inhibiting the expression of CIP2A can reduce the proliferation,invasion and migration ability of glioma cell lines.3.Inhibiting the expression of CIP2A can attenuate the EMT-like progression of glioma cell lines.
