Objectives:To detect the expression of PDCD4 gene protein in cancer tissues and normal tissues adjacent to cancer in patients with penile cancer,analyze the difference of the expression of PDCD4 protein in the penile cancer and its paired para-carcinoma tissues,investigate the role and significance of PDCD4 gene in penile cancer.and to explore its potential as a diagnostic marker and therapeutic target,the penile cancer tissue and adjacent tissue were analyzed.Methods:(1)In this study,a total of 9 penile cancer patients who were admitted to the urology department of The Second Affiliated Hospital of Kunming Medical University from May 2019 to December 2019 and met the study conditions were selected as clinical specimens.Penile cancer tissues(study group)and adjacent normal tissues(away from the tumor>2cm)of each patient were collected by means of self-matching(control group).The samples were divided into three parts.A part of the specimen was stored in a PE tube containing two-thirds volume of RNA-Later preservation solution,those samples were put into a liquid nitrogen container to freeze immediately,and then stored at-20? for later use for genetic extraction and detection;A part of the specimen were washed with normal saline and placed in a clean sterile PE tube and kept in a-70? liquid nitrogen tank for Western blotting(WB)experiments.The remaining one-third of the specimens were placed in formalin fixative solution and stored in a refrigerator at 4? for immunohistochemical detection(IHC).(2)qRT-PCR technology was used to determine the expression of PDCD4mRNA in penile carcinoma and its adjacent tissues.WB technique was used to determine the expression of PDCD4 protein in penile carcinoma and its adjacent tissues.IHC technique was used to determine the expression and cell localization of PDCD4 protein in penile carcinoma and its adjacent tissues.(3)SPSS 19.0 statistical software was used for data statistics and analysis.The measurement data was expressed as mean ± standard deviation(x ± s).Paired t test was used for comparison within the group,and P<0.05 was considered statistically significant.The brown-yellow of IHC was defined positive.The statistical graph was drawn with GraphPad Prism 9.0 software.Results:1.Real-time fluorescence quantitative reverse transcription polymerase chain reaction(qRT-PCR)to detect the relative expression of PDCD4 mRNA in cancer and adjacent to cancerThe quantity of PDCD4mRiNA in 9 cases of penile cancer tissues and their adjacent tissues showed that the relative expression of PDCD4 mRNA in penile cancer tissues and adjacent tissues was significantly increased,the difference.was statistically significant(3.327±0.482,0.185±0.036;P<0.05).2.Western blot(WB)to detect the relative expression of PDCD4 protein in cancer and adjacent to cancerThe results of the quantity of PDCD4 protein in 9 cases of penile cancer tissues and their adjacent tissues showed that the relative expression of PDCD4 protein in penile cancer tissues were more than the adjacent tissues,the difference was statistically significant(1.047 ± 0.084,0.517±0.083;P=<0.05).3.Immunohistochemistry(IHC)to detect the relative expression of PDCD4 protein in cancer and adjacent to cancerThe immunohistochemical staining of quantitys of PDCD4 protein in 9 penile carcinoma tissues and their paired paracancerous tissues showed that the penile cancer tissues were more than the adjacent tissues,the value number was 26.540 ± 1.978,3.308±0.350 respectively,P<0.05.The brown-yellow were seen in the nucleus and cytoplasm on li ght microscopeConclusions:1.PDCD4 positive is brownish yellow,located in the nucleus,cytoplasm2.The expression of PDCD4 in penile cancer tissues were significantly higher than that in normal paracancerous tissues,indicating that PDCD4 might be associated with the occurrence of penile cancer.3.The expression of PDCD4 were up-regulated at transcription and translation levels and consistent in both penile carcinoma tissues and paracancerous tissues.
