Objective:Cisplatin is a common chemotherapeutic agent for tongue squamous cell carcinoma,but drug resistance is the main limiting factor for its clinical treatment.The potential mechanism of cisplatin resistance in tongue squamous cell carcinoma has not been fully elucidated.This topic is to study the role and mechanism of LAMP2 in cisplatin resistance of tongue squamous cell carcinoma cells,and to provide a new strategy for the treatment of clinical drug resistance of tongue squamous cell carcinoma.Methods:Cisplatin resistant cells of tongue squamous cell carcinoma were established by repeated induction of low dose cisplatin;Western blot was used to detect the expression level of autophagy-related proteins after cisplatin treatment of tongue squamous cell carcinoma.;MTT assay was used to detect the growth inhibition of tongue squamous cell carcinoma cells by different concentrations of cisplatin and to determine the concentration of the selected drug;Expression levels of autophagy-related proteins after cancer cells;SiRNA was used to knock down the expression of LAMP2 in tongue squamous cell carcinoma cells,MTT assay and AnnexinV-FITC/PI flow cytometry was used to detect the apoptosis-inducing effect of drugs in tongue squamous cell carcinoma.Results:The cisplatin-resistant human tongue squamous cell carcinoma cell line SCC 25/DDP was successfully constructed.After cisplatin treated SCC25/DDP,the expression of LAMP2 increased gradually with the prolongation of treatment time,but the expression of LAMP2 in parent SCC25 cells decreased gradually Chloroquine reverses cisplatin resistance in tongue squamous cell carcinoma;Knocking down LAMP2 increased the sensitivity of tongue squamous cell carcinoma to cisplatin.Conclusions:Cisplatin induced the increase of LAMP2 expression in tongue squamous cell carcinoma cell line,autophagy induced cisplatin resistance in tongue squamous cell carcinoma cell line.Knocking down LAMP2 expression enhanced cisplatin sensitivity,inhibited proliferation and promoted apoptosis of tongue squamous cell carcinoma cell line.This study provides a new idea and direction for solving the problem of cisplatin resistance in clinical tongue squamous cell carcinoma.
