Objective:By establishing a soft tissue healing model for rabbit tooth ext raction,the effects of platelet-rich fibrin on promoting soft tissue healing in ra bbit tooth extraction were histologically investigated.Method:Extraction of the left and right maxillary central incisors of 18 Japanese large white-ear rabbits to establish a model of wound healing,and the left and right extraction wounds were compared with each other.The left extraction was the experimental group.Platelet-rich fibrin was placed in the left alveolar socket and sutured while the right extraction was used as the control group.After the blood clot filled the alveolar socket by itself,the right extraction was sutured.On the 3rd,5th,7th,11th,and 14th days after surgery,the healing of the soft tissues of the experimental group and the control group were visually observed,and the soft tissue specimens of the tooth extractions were collected after sacrifice on the 3th,7th,and 14th days after the operation.HE staining was used to observe the morphology.Immunohistochemical staining was used to observe the expression of TGF-?1 and?-SMA in the soft tissues of gingival extraction wounds.The average photometric value of the tissue specimens was analyzed by Image J software.The obtained experimental data were analyzed statistically.Using t test to compare whether there is statistical difference between the two groups(P<0.05).Results:On the 3th day after surgery,the gums in the experimental group were not noticeably red and swollen,while the control group showed obvious swelling and swollen.The HE staining results showed that compared with the control group,obvious angiogenesis was observed in the lamina propria of the soft tissue of the extraction group,and new fibroblasts were seen around the blood vessels.On the 7th day after surgery,the HE staining results showed that the number of angiogenesis and collagen fibers in the experimental group were significantly higher than those in the control group.On the 14th postoperative day,HE staining results showed that a large number of neovascularizations were formed in the experimental group,and the collagen fibers were thick and well-ordered.On 3th,7th,and 14th days after surgery,immunohistochemical staining showed that the expression of TGF-?1 and ?-SMA protein in the experimental group was stronger than that in the control group,and the difference was statistically significant(P<0.05).Conclusion:Platelet-rich fibrin may increase the expression of TGF-?1 and?-SMA,thereby promoting the healing of soft tissues during tooth extraction.
