Objective:to investigate the changes of serum A-kinase anchoring protein 1(AKAP1)in people suffering type 2 diabetes mellitus(T2DM)and obesity,the influencing factors of AKAP1,and the correlation between serum AKAP1 and the incidence of T2DM and obesity.Methods:A total of 261 people were involved in the experiment,including 130 patients with newly diagnosed T2DM and 131 people with normal glucose tolerance(NGT).According to the results of body mass index(BMI),they were further divided into four groups.People with T2DM but normal weight(NW)were assigned to T2DM+NW group,with T2DM and overweight(ow)or obesity(OB)assigned to T2DM+OW/OB group,and the like,NGT+NW group,NGT+OW/OB group.The serum level of AKAP1 was detected by ELISA method,and compared by one way ANOVA.Pearson correlation was used to study the correlation between AKAP1 and other variables,linear regression was adopted to determine the independent factors of AKAP1.Multiple logistic regression was used to analyze the relationship between AKAP1 with T2DM and obesity.Results:the serum AKAP1 level in NGT+NW group was 1.74 ± 0.42ng/ml,higher than that in NGT+OW/OB group,vs 1.59± 0.41ng/ml,P<0.05,and T2DM+OW/OB group,vs 1.52 ± 0.36ng/ml,P<0.01.In this experiment,the AKAP1 level of 130 subjects with T2DM was lower than that of NGT population,1.57 ± 0.35ng/ml vs 1.67± 0.42ng/ml,P<0.05.Serum AKAP1 level of 127 OW/OB people was lower than that of NW people,1.55 ± 0.38ng/ml vs 1.68 ± 0.39ng/ml,P<0.01.Pearson correlation showed that in NGT population,AKAP1 was negatively correlated with BMI,diastolic blood pressure(DBP),waist hip ratio(WHR)and triglyceride(TG),r values were-0.182,-0.201,-0.203 and-0.192,respectively.In patients with T2DM,AKAP1 was negatively related to BMI,systolic blood pressure(SBP),fasting insulin(FINS)and insulin resistance(IR),r values were-0.224,-0.175,-0.188 and-0.202,respectively.Linear regression showed that BMI,DBP and IR were independent factors related to AKAP1,?=-0.032,-0.006 and-0.019,respectively,while hip circumference(HC)might be positively correlated with AKAP1,?=0.010.According to AKAP1 levels,the subjects were divided into three groups.Compared with highest tertile,the prevalence of T2DM was higher in the other two groups,OR was 2.207(95%CI 1.203,4.050)and 2.051(95%CI 1.121,3.753),respectively;and the incidence of obesity in lowest tertile was higher,OR was 3.009(95%CI 1.626,5.568).The area under ROC curve of using serum AKAP1 predicting T2DM and OB/OW was 0.421 and 0.377 respectively.Conclusions:serum AKAP1 level of people suffering diabetes or obesity was slightly decreased.Serum AKAP1 may be negatively correlated with DBP,BMI and IR.The risk of type 2 diabetes and obesity was increased in people with lower levels of serum AKAP1.It is of small significance that using serum AKAP1 to evaluate T2DM or obesity.
