Euchresta tubulosa Dunn is a kind of legume plant,which has the functions of anti-inflammatory and so on.It is used to treat stomach and abdominal pain,sore throat and so on.It is one of the widely used and high-efficiency drugs for Tujia people and other ethnic minorities in Wuling mountain area.At present,there are few reports on the chemical constituents of Euchresta tubulosa Dunn at home and abroad,among which alkaloids are of great research value.In this paper,the preparation,structure identification and activity test of alkaloid monomers in the rhizome of Euchresta tubulosa Dunn were studied.The main contents are as follows :? The total alkaloids in the roots and stems were extracted by microwave temperature control and ultrasonic wave.The results showed that the extraction time was 37.8 min,the microwave heating temperature was 64.5 ?,the ultrasonic power was 575 W,the ethanol volume fraction was 95 %,and the total alkaloid yield was 0.5436 %,which was higher than 0.2436 % of no microwave-assisted ultrasound,0.4538 % of sonication-assisted,0.3884 % of ethanol reflux;It was found that the content of total alkaloids in roots and stems was higher than that in leaves,which could be used as medicinal parts.? The active components in the rhizome of Euchresta tubulosa Dunn were extracted by ethanol thermal reflux,and the total alkaloids were enriched by chloroform solvent extraction.Four components were separated by silica gel column chromatography.The different components were separated and purified by HSCCC method,and five kinds of monomers were finally obtained.? After the purity analysis of the isolated monomers by HPLC-UV,five monomers were identified by NMR,MS and other spectroscopic methods,which were matrine,Oxymatrine,N-formyl-cytisine,N-acetyl-cytisine and cytisine.Among them,N-formyl cytisine is the first one isolated from the plant.? The traditional preparation method of alkaloids in the roots of guanyushan bean has a long cycle and complicated steps,so the exploration of efficient,economic and environmental protection preparation methods can provide a material basis for related research.The model of separation and purification of the monomers has been optimized.The model of liquid-liquid extraction and HSCCC gradient elution has been established.Four alkaloids,matrine,Oxymatrine,N-formyl cytisine and N-acyl cytisine,can be prepared at the same time;All solvents in the instrument were blown out with nitrogen,it is found that the content of cytisine in the lower phase is very high.Finally,the effects of the velocity of the mobile phase and the rotating speed of the spiral string on the retention of the stationary phase were studied.It is found that the model has wide applicability.? In vitro antioxidant test was carried out on the total alkaloids enriched in the rhizome of Phaseolus angustifolia.It was found that the alkaloids had certain scavenging effect on free radicals.The activity of the three alkaloid monomers was tested by the methods of azurol and CCK8.
