ObjectTo explore the effect and potential pathway of Aluminum magnesium sulfate in the treatment of acetic acid-induced gastric ulcer in rats.Methods1.Adult male SD rats were randomly divided into control group,model group,Aluminum magnesium sulfate low dose group,middle dose group and high dose group.The gastric ulcer was established by acetic acid etching.Different doses of Aluminum magnesium sulfate were given for intervention.The gastric mucosal morphology was observed and the ulcer area was calculated.Then we detected the apoptosis of gastric mucosal cells by TUNEL staining,detected the expression of ERS-related molecules and growth factors in gastric mucosa by Real-time PCR.2.Human gastric mucosal epithelial cells(GES-1)were divided into control group,acetic acid group,Aluminum magnesium sulfate pretreatment group and drug group.Acetic acid was used to make model,Aluminum magnesium sulfate were given for intervention.Then we detected the cell viability by MTT method,detected the expression of ERS-related protein of cells by Western Blot.Results1.The gastric ulcer model in rats is established by acetic acid etching.Aluminum magnesium sulfate can effectively reduce the ulcer area and promote ulcer healing.2.Acetic acid stimulation can significantly increase the expression of ERS-related molecules IRE1?,PERK,GRP78 and CHOP mRNA in gastric mucosa,and then induce the apoptosis of gastric mucosal cells.Aluminum magnesium sulfate can reduce the expression of IRE1?,PERK,GRP78 and CHOP mRNA significantly,and alleviate the apoptosis of gastric mucosal cells.Aluminum magnesium sulfate can significantly increase the expression of VEGF and EGF mRNA in gastric mucosa.3.Acetic acid stimulation inhibit the proliferation of GES-1 cells in a concentration dependent manner.Aluminum magnesium sulfate can effectively reduce the injury of GES-1 induced by acetic acid.4.Acetic acid stimulation can significantly increase the phosphorylation level of p-IRE1? and p-eIF2?,increase the expression of ATF6 and CHOP in GES-1 cells.Aluminum magnesium sulfate can effectively decrease the phosphorylation level of p-IRE1? and p-eIF2?,decrease the expression of ATF6 and CHOP in GES-1 cells.ConclusionsAluminum magnesium sulfate can treat gastric ulcer effectively,the possible pathway is inhibiting endoplasmic reticulum stress to reduce the apoptosis of gastric mucosal cells,up-regulating the expression of growth factors to enhance the ability of mucosal repair.
