Objective:To detect the expression of RAD51 and XRCC4 proteins in esophageal squamous cell carcinoma cell line TE-1,ECA109,normal esophageal epithelial cell line HEEC,and paraffin-embedded esophageal squamous cell carcinoma tissues(including their matched cancer metastatic lymph node tissues),and to discuss the effect of RAD51,XRCC4 protein on the occurrence,development and prognosis of esophageal squamous cell carcinoma.Materials and methods:1.Culture and passage of esophageal squamous cell line TE-1,ECA109 and normal esophageal epithelial cell line HEEC.2.Immunocytochemical method was used to detect the expression of RAD51 and XRCC4 proteins in esophageal squamous carcinoma cell lines TE-1,ECA109 and normal esophageal epithelial cell line HEEC.3.Immunohistochemical method was used to detect the expression of RAD51,XRCC4 protein in 62 cases of esophageal squamous cell carcinoma and cancer tissues in lymph nodes and 14 cases of adjacent cancer tissues.4.Use SPSS 17.0 statistical software to complete data statistics(P<0.05 was considered as statistically significant);Chi-square test for comparison between groups;Spearman rank correlation test for correlation analysis;Kaplan-meier draw survival curves.Results:1.RAD51 protein and XRCC4 protein were positively expressed in esophageal squamous cell line TE-1 and ECA109,and negatively expressed in normal esophageal epithelial cell line HEEC.2.RAD51 protein was positively expressed in esophageal squamous cell carcinoma tissues.Compared with the adjacent tissues,the positive expression rate was up-regulated,with a statistically significant difference(P=0.000).However,there was no statistically significant difference between the positive expression rate of RAD51 protein and that of the cancer tissues in the paired lymph nodes(P=0.065).XRCC4 protein was positively expressed in esophageal squamous cell carcinoma tissues.The positive expression rate was up-regulated compared with the adjacent tissues(P=0.041).The positive expression rate was up-regulated compared with the cancer tissues in paired lymph nodes(P=0.007).3.The positive expression rates of RAD51 and XRCC4 protein were not correlated with other clinicopathological characteristics(all P values were>0.05).Only the positive expression rates of RAD51 protein were correlated with different gender.4.There was no correlation between the positive expressions of RAD51 and XRCC4 protein in the included samples.The difference was not statistically significant(r=0.158,P=0.219).5.There was no significant difference in progression-free survival(PFS)between patients with positive RAD51 protein expression(P=0.866).There was no significant difference in PFS between patients with positive XRCC4 expression(P=0.106).In addition,there was no significant difference in PFS between RAD51 protein-positive patients and XRCC4 protein-positive patients(P=0.644).Conclusion:RAD51 protein and XRCC4 protein may be involved in the occurrence of esophageal squamous cell carcinoma by participating in DNA double-strand breaks(DSBs)repair and play a role in lymph node metastasis,but their positive expression of enrolled specimens is not related to the prognosis of patients.Further experiment is needed to verify the relationship between RAD51 and XRCC4 and esophageal squamous cell carcinoma.
