Objective:To explore the effects of SCR-1 in cerebral cortex and hippocampal on the expression of synaptic protein PSD95 and whether SRC-1 is one of the molecular mechanisms of synaptic regulation by observing the changes in the expression of PSD-95 in the cerebral cortex and hippocampus after lipopolysaccharide(LPS)up-regulation and specific plasmid down-regulation of SRC-1 expression in cerebral cortex and hippocampus.This experiment will provide a basis for further study on the mechanism of SRC-1 brain function.Methods:Male Kunming mice were randomly divided into four groups,8 mice in each group.Saline,LPS,specific plasmid,and specific plasmid+LPS were added to interfere with the expression of SRC-1 and cortical and hippocampal tissues of mice excised after four different interventions.Then we test the LPS-induced inflammation level at different times by ELISA and the SRC-1 levels after adding LPS and specific plasmid intervention by Western Blot.Finally,Western Blot and immunohistochemical techniques were used to observe changes in SRC-1 expression levels and corresponding PSD95 expression levels in the cerebral cortex and hippocampus of mice.Results:1.ELISA results showed that the expression levels of IL-17 and TNF-?were highest at 12 h;2.Western Blot results showed that the expression level of SRC-1 was the highest at 12 hours after LPS injection,and the expression level of SRC-1-mus-2401 was the lowest among the four gene loci after specific plasmid injection;3.The results of Western Blot and immunohistochemistry showed that when compared with the normal saline group,the SRC-1 expression level in the LPS group was significantly increased and the PSD95 expression level was significantly decreased;the SRC-1 expression level in plasmid group was significantly decreased and the PSD95 expression level was significantly increased;the expression level of SRC-1 decreased and the expression level of PSD95 increased in the plasmid group+LPS group.Conclusions:1.SRC-1 in the cerebral cortex and hippocampus has a significant effect on PSD95 expression levels,and there is a negative correlation between them,suggesting that SRC-1 is one of the molecular mechanisms of synaptic regulation;2.SRC-1 is widely expressed in the brain,suggesting that SRC-1 is involved in the regulation of a variety of brain functions,and we will investigate more in the future. |
