| ABSTACTMyofascial pain syndromes(MPS),also known as myofascial pain,refers to pain caused by chronic aseptic inflammation of muscles and their fascia.It is a common cause of neck,shoulder,waist and leg pain.Myofascial trigger points(MTrPs)are specific symptoms,manifested as hard scattered muscle tension bands in skeletal muscle,which usually cause spontaneous pain and can also induce pain when under pressure.The main pathological changes of myofascial pain syndromes are the increase of inflammatory factors and pain transmitters in the lesions,Eventually developed into chronic aseptic inflammation of muscles;muscle satellite cells(MuSC)are the cells of repairing damaged muscle,which closely linked to local inflammatory states.Sympathetic nerves can not only affect both the reconstruction of skeletal muscle in MPS individuals and the inflammatory of muscles.However,the role of sympathetic nerves and muscle satellite cells in the MPS has not been confirmed by relevant studies.Objective:To investigate whether sympathetic nerves affect the Proliferation of muscle satellite cells in MPS sufferers,and whether sympathetic nerves and muscle satellite cells can synergistically affect the local and overall inflammaion of MTrPs Rethod:1.Thirty rats were randomly divided into 5 groups:injection control group(group A);experimental group(group B),injection combined with blow group(group C),blow control group(group D),nerve destroy group(group E)).Modeling in two stages:group A:injection of normal saline+normal feeding;B:sympathetic nerves destroying+MPS modeling in left legs;C:injection of normal saline+MPS modelin in left legs;D:normal feeding+MPS modelin in left legs;E:sympathetic nerves destroying+ normal feeding.electromyography and palpation were used to filter successful models.2.Measure the skin temperature of MTrPs and the same part on the opposite side at different time periods before and after the MPS model is built.3.Frozen slices were made and HE staining was performed to observe the muscle fiber morphology and inflammation status.The cross-sectional area of muscle fibers was measured and the inflammatory cells count was used to assess the local inflammatory status.4.Rat blood was centrifuged to obtain rat plasma,and the concentrations of 5-HT,NE and TNF-α in rat plasma were measured by ELISA.5.Take the quadriceps muscles of the rats,measuring the expression levels of MyoG and MyoD genes in the quadriceps muscles of the rats by Realtime-PCR.Result:1.There was no significant difference in skin temperature between rats in groups A and E.The skin temperature of rats in groups A and E was significantly lower than that of rats in groups B,C,and D in the second measurement.The skin temperature of the rats in group B was significantly higher than that in the other groups at 1 day after the first MPS modeling,and the skin temperatures of the other groups were not significantly different.The body temperature of the rats in group B was significantly higher at the 1 day after the last MPS modeling.The body temperature of rats in groups A,C and E was not significantly different in the other groups.There was no significant difference in body temperature between the groups at the first、forth、fifth and seventh measurement.2.Observation under the light microscope shows that the muscle fibers of the skeletal muscles of the A and E groups are arranged regularly and neatly,and the muscle space is uniform.There is no inflammatory cell infiltration.The muscle fibers of the E group are slightly atrophic than the A group;The cross section of the muscle fibers in the group was reduced to varying degrees,the muscle fiber gaps were widened and the sizes were different,the muscle fibers were arranged disorderly,and different degrees of breaks and distortions appeared.The inflammation of the connective tissue around and around the muscle fibers in group B was the most severe.3.The count of inflammatory factors in the left muscle slices of rats in groups B,C,and D were significantly higher than those in the contralateral side.There was no significant difference in the count of inflammatory factors in bilateral muscle slices of rats in groups A and E.The count of inflammatory factors in the left muscle section of the rats in group B was significantly higher than that in the other groups,and the count of inflammatory factors in the left muscle sections of C and D rats was significantly higher than in the groups of A and E.There was no significant difference in inflammatory factor counts.The count of inflammatory factors in the right muscle sections of group B rats was significantly higher than that in groups A,D,and E,while the concentrations of inflammatory factors in the right muscle sections of groups A,D,and E were not significantly different.B,C,and C,There was no significant difference in inflammatory factor counts in right muscle sections between groups D.4.There was no difference in the average TNF-α concentration in the plasma of rats in different groups;the NE concentration in the plasma of group A was greater than that in the other groups,and there was no difference in the NE concentration in the plasma of the other groups;5HT in the plasma of rats in groups A and D The concentration was higher than that in groups B and E.The concentration of 5HT in the plasma of rats in group B was less than that in groups C and D.The concentration of 5HT in the plasma of rats in group E was less than that in group D.5.The Myog expression folds in the left thigh muscles of group A rats is greater than that of B and C groups;the MyoD expression folds in the bilateral muscles of group A rats is greater than that of group E rats;the MyoD expression folds in the left muscles of group C rats is greater than D,E rats.The MyoD expression folds in the right muscle of group A rats was greater than that of rats in groups B,D,and E.There was no significant difference in the MyoD expression folds in the contralateral muscles of any group of rats.Conclusion:1.Destroying the sympathetic nerve can cause a temporary rise in local skin temperature at the trigger point;2.Destroying sympathetic nerves can affect growth of muscle fiber and affect the inflammation of MTrPs and plasma;3.Destroying the sympathetic nerve has no effect on the Proliferation of muscle satellite cells at the trigger points;4.It can not be confirmed that sympathetic nerves can affect the inflammation of rats by affecting the Proliferation of muscle satellite cells. |
