Aims Study on the anti-laryngeal cancer activity of Dehydrocostus lactone monomer compounds extracted from Saussurea costus,and explore the possible molecular mechanism of dehydrocostuslactone against laryngeal cancer.Methods Dehydrocostus lactone was extracted from Saussurea costus by high performance liquid phase separation,and cell culture technology,CCK-8 method,Hoechst33258 staining method,Flow cytometry technology,Western Blot experiment,Immunofluorescence,Immunohistochemistry Other experimental methods to analyze the anti-laryngeal cancer activity of Dehydrocostus lactone and its intrinsic molecular mechanism.In animal experiments,BALB / C nude mice were injected subcutaneously with a certain amount of Hep-2 cells to establish a Hep-2 tumor-bearing nude mouse experimental model,and Dehydrocostus lactone was introduced into nude mice by intraperitoneal injection.In vivo,continuous administration for four weeks,and the size of solid tumors and related experimental detection of protein in tumor tissuesResults Dehydrocostolactone was isolated from radix aconitae and was combined with pharmacologic experiments.It was found that dehydrocostolactone could inhibit the proliferation and migration of laryngeal cancer cells in a dose-time dependent manner.In addition,at a certain dose,Dehydrocostus lactone is less cytotoxic to normal laryngeal epithelial cells.Dehydrocostus lactone induced apoptosis in laryngeal cancer cells in a dose-dependent manner.The results of Western blots,Immunohistochemistry,and Immunofluorescence experiments on Dehydrocostus lactone against laryngeal cancer showed that Dehydrocostus lactone exerted anti-laryngeal cancer effects through the PI3 K / Akt / Bad signaling pathway;Dehydrocostuslactone up-regulates the protein expression of the apoptotic gene Bax,down-regulates the protein expression of the apoptotic gene Bcl-2,and activates Caspase-3,9,12 and promotes the expression of Grp78 and CHOP to activate the intrinsic apoptosis pathway Thereby inducing apoptosis in laryngeal cancer cells.Conclusion Dehydrocostus lactone can inhibit the proliferation and migration of laryngeal cancer cells,and is less toxic to normal laryngeal epithelial cells.In vivo and in vitro experimental studies have shown that Dehydrocostus lactone can induce laryngeal cancer cell apoptosis by regulating the PI3K/Akt/Bad signaling pathway,activating the mitochondrial apoptosis pathway and stimulating the endoplasmic reticulum stress response,so as to achieve the anti-laryngeal cancer effect.
