Aging is an inevitable process and a spontaneous process caused by many factors.As the global aging problem becomes more and more serious,research on aging and interventions related to aging is receiving increasing attention.Because clinical trials are too long and there are some ethical issues,animal models are necessary for aging research.As one of the key model animals for aging,C.elegans has been widely used.Lycium barbarum polysaccharide is a natural product,it has been widely reported having various functions such as regulation of immunity and antioxidant effects.In this paper,Caenorhabditis elegans as a model organism,many aspects of the life and health barbarum polysaccharides were evaluated.Then the molecular signaling pathways were then analyzed by nematode mutants,RNAi and RT-PCR.We studied the lifespan of C.elegans cultured at different concentrations of LBP and found that LBP could extend lifespan.The extending lifespan was not simply linearly dependent on the dose of LBP,and it was very interesting that there was an optimal concentration of 300 ?g/ml.The lifespan extension effect of LBP on worms did not impact by the quality of dietary intake,and it was independent of temperature and non-heritable.LBP was also found to enhance the viability of N2 under high temperature,strong oxidation and heavy metals.LBP promotes the antioxidant capacity of the worms,significantly increases the activity of CAT and SOD to scavenge oxidative free radicals,and suppresses the detrimental effect of ROS,thereby reducing oxidative damage of cells and improving the health.LBP can also enhance on the reproductive potentials and muscle integrity of C.elegans.In a word,LBP did not only extend lifespan but also improve health status of the nematodes.Many genetic pathways have been shown to be conserved,and it is necessary to elucidate the molecular mechanisms by which LBP extends lifespan.Sir-2.1 and daf-16 are extremely outstanding genes in the ?S pathway,as well,DAF-16/FOXO transcription factors are also key components of the IIS pathway associating with aging and metabolism.We found that LBP has a shorter extending lifespan effect on mutants of sir-2.1 and daf-16 than on N2.This means that LBP extended lifespan requires sir-2.1 and daf-16.For daf-16 RNAi,there is no difference in the effect of LBP on N2 and sir-2.1 mutants,which implies that sir-2.1 may play a role dependent of daf-16.The daf-12 gene plays an important role in aging,immunity and antioxidation.We found that LBP had a shorter extending lifespan effect on daf-12 mutants than on N2,implying that LBP extended lifespan requires daf-12.daf-16 RNAi was done to the daf-12 mutants and daf-12 RNAi was done to the daf-16 mutants,and it showed that LBP lost the function of prolonging lifespan,which suggests the effects of LBP on aging of C.elegans depend on daf-12 and daf-16.In summary,LBP has the functions of prolonging the lifespan,maintaining the environmental adaptability,reproductive capacity and motility of C.elegans.The effects of LBP on the health and aging of C.elegans are regulated by sir-2.1,daf-12 and daf-16.Our study the role of LBP in aging and health,and its molecular mechanism.It also provided a basis for further research on LBP and LBP as a food or drug for intervention of aging.
