ObjectiveAnalysis of the expression of ARHGAP 17 and selective splicing variants in hepatocellular carcinoma and paracancerous tissues.By discussing the correlation between ARHGAP 17 and different selective shear variants and tumor stage and prognosis,the selective shear variants of ARHGAP 17 which play a major role in different stages of hepatocellular carcinoma were determined.The effects of different factors on the prognosis of patients after hepatectomy were further analyzed.MethodsTissue samples were collected from 31 patients who underwent hepatectomy in the Department of Hepatobiliary surgery of General Hospital of Northern Theater Command from June 2015 to September 2016.The deadline for follow-up is August 2019.All the patients with liver cancer did not receive any treatment before hepatectomy,and all of them were diagnosed as hepatocellular carcinoma by pathology.The patients were followed up by outpatient clinic and telephone.The content of follow-up was the overall survival time and disease-free survival time of the patients.The expression of ARHGAP 17 and selective shear variants(ARHGAP 17-S,ARHGAP 17-L)in 31 pairs of hepatocellular carcinoma tissues and paracancerous tissues were detected by RT-PCR.To analyze and observe the following points: 1.whether there is a difference in the expression of ARHGAP 17 between hepatocellular carcinoma and paracancerous tissues;2.Whether there are differences in the expression and distribution of different alternative splicing variants of ARHGAP 17 between hepatocellular carcinoma and paracancerous tissues;3.The clinical data of patients with hepatocellular carcinoma were retrospectively analyzed to explore the correlation between the expression of ARHGAP 17 and its different selective splice variants and prognosis.Graphpad Prism 8.0 and SPSS 17.0 statistical software were used to process and analyze the related data.The measurement data are expressed by the mean ± standard deviation(X? ± ).The comparison between groups was analyzed by student's test or ANOVA analysis of variance.The counting data were tested by ? 2 test.The survival of the patients was evaluated by Kaplan-Meier method.Log-rank test was used to compare and analyze the overall survival rate(OS)and disease-free survival rate(DFS)among different groups.Then the statistically significant indicators were analyzed by multivariate Logistic regression analysis to further determine the independent risk factors affecting the prognosis after hepatectomy.The effects of various factors on the overall survival time and disease-free survival time of patients were observed.P< 0.05 indicates that it is statistically significant.Results1.The RT-PCR results show that: The minimum expression of ARHGAP 17-S in(T)was 0.864,the maximum expression was 2.102,and the average expression was 0.664 ±0.490.The minimum expression of(NT)in paracancerous tissues was 0.237,the maximum expression was 1.963,and the average expression was 1.000 ±0.477.The relative expression of ARHGAP 17-S in cancer tissues was significantly lower than that in paracancerous tissues.2.There was a significant difference in the relative expression of ARHGAP 17-S between stage 1 and stage 2-3 HCC tissues,and between stage 1 liver cancer tissues and paracancerous tissues(P<0.01).There was a significant difference in the relative expression of ARHGAP 17-S between stage 1 HCC tissues and paracancerous tissues(P<0.05).3.The survival time of OS and DFS in patients with high expression of ARHGAP 17-Total was longer than that in patients with low expression of patients(P<0.05).4.In the paracancerous tissues of patients with stage 2-3 HCC,the OS of patients with high expression of ARHGAP 17-S was shorter(P<0.05);In the paracancerous tissues of patients with stage 1 HCC,the patients with high expression of ARHGAP 17-S had longer time of DFS(P<0.05).In the paracancerous tissues of patients with stage 2-3 HCC,the DFS of patients with low expression of ARHGAP 17-S was longer(P<0.01).5.The tumor diameter,tumor location,AJCC stage,BCLC stage,AFP level,intraoperative blood loss and operation time were the risk factors affecting the prognosis of HCC.The results of univariate analysis were analyzed by Logistic multivariate regression analysis.The results showed that the length of operation time was an independent risk factor for the prognosis of patients after hepatectomy.ConclusionsThe relative expression levels of ARHGAP 17-Total and ARHGAP 17-S play an important role in the occurrence and development of HCC.The postoperative survival time of patients with high expression of ARHGAP 17-Total was longer.ARHGAP 17-S plays an indicative role in predicting the recurrence of HCC.As an independent risk factor for the prognosis of HCC,the length of operation is closely related to the prognosis of patients.
