Objective:To Determine Whether There Are Differences In The Expression Of VEGF And FGF-2 between cholesterol polyp and normal gallbladder wall submucosa.Furthermore,the expression of VEGF and FGF-2 and their differences were discussed to determine whether there was a correlation between the occurrence and growth of cholesterol polyps.Method:from October 2018 to October 2019,40 pathological wax specimens of patients with cholesterol polyps who underwent cholecystectomy in the second Hospital of Chifeng City from October 2018 to October 2019 were collected and tissue sections were made.The characteristics of tissue and cell structure of cholesterol polyps were observed under HE staining microscope.The sections of cholesterol polyps and their surrounding normal gallbladder walls were stained with VEGF and FGF-2 respectively by immunohistochemical staining technique.The expression and characteristics of VEGF and FGF-2 in the two groups were observed under microscope.The optical density of VEGF and FGF-2 expression in each group was measured by IPP6.0 image analysis software.SPSS 21.0 software was used for statistical analysis to determine whether there was a difference in the expression of the two factors between the two groups.Results:1.HE staining observation:all cholesterol polyps were semicircular protuberances or finger-like protuberances protruding to the gallbladder cavity under the microscope,covered by intact mucosal epithelial cells,and polyps contained a large number of foam-like macrophages that engulfed cholesterol,during which there were connective tissue and capillaries,and the pedicle capillaries were rich.Compared with the surrounding normal gallbladder wall submucosa,it was found that the capillary density was relatively higher,and a small amount of cholesterol deposition could be seen in part of the gallbladder wall submucosa.2.The expression of VEGF in normal gallbladder wall and cholesterol polyps:VEGF was expressed in mucosal epithelial cells,fibroblasts and capillary endothelial cells of normal gallbladder wall,especially in capillary endothelial cells and mucosal epithelial cells.VEGF was expressed in mucosal epithelial cells,capillary endothelial cells,foam cells and fibroblasts in cholesterol polyps,especially in capillary endothelial cells,mucosal epithelial cells and foam cells.The results showed that the expression intensity of VEGF in cholesterol polyp group was higher than that in normal gallbladder wall group.IPP6.0 image analysis software showed that the average optical density of VEGF expression in normal gallbladder wall group was 0.04 ±0.02,and the average optical density of VEGF expression in cholesterol polyp group was 0.1±0.03.Compared with normal gallbladder wall group,the expression of VEGF in cholesterol polyp group was significantly higher than that in normal gallbladder wall group(P<0.05).3.The expression of FGF-2 in normal gallbladder wall and cholesterol polyps:FGF-2 was expressed in mucosal epithelial cells,fibroblasts and capillary endothelial cells of normal gallbladder wall,especially in fibroblasts and mucosal epithelial cells.FGF-2 was expressed in mucosal epithelial cells,capillary endothelial cells,foam cells and fibroblasts in cholesterol polyps,especially in fibroblasts,mucosal epithelial cells and foam cells.The results showed that the expression intensity of FGF-2 in cholesterol polyp group was higher than that in normal gallbladder wall group.IPP6.0 image analysis software showed that the average optical density of FGF-2 expression in normal gallbladder wall group was 0.04±0.01,and that in cholesterol polyp group was 0.1±0.02.Compared with the normal gallbladder wall group,the expression of FGF-2 in cholesterol polyp group was significantly up-regulated,and the difference was statistically significant(P<0.05).3.Conclusion:1.There was significant difference in the expression of VEGF and FGF-2 between cholesterol polyp and normal gallbladder wall,and the expression in cholesterol polyp group was significantly higher than that in gallbladder wall group(P<0.05).2.There is a correlation between VEGF and FGF-2 in the occurrence and growth of cholesterol polyps,which may play a promoting role.
