Objective:(1)To optimize the extraction and preparation process of Mongolian medicine Jidou antitussive granules.(2)Evaluate the anti-cough,expectorant,antiasthmatic and anti-inflammatory pharmacological effects of Jidou antitussive granules.Methods:(1)The extraction process of Jidou antitussive granules was optimized through single factor inspection and orthogonal experiment,Using the extraction rate,the content of liquiritin and Ammonium Glycyrrhizate as indicators.(2)Screening the best auxiliary materials,auxiliary material ratio,wetting agent and binder to determine the best prescription rate,using the single factor inspection,based on the granulation situation,particle size and moisture absorption percentage.(3)In the main efficacy experiment,animals were divided into normal group,model group,three dosage groups of Jidou antitussive granules and three dosage groups of Jidou Zhike San(low,medium and high dosage).Observe the cough incubation period and the number of coughs of mice through ammonia cough test;examine the expectorant effect through mouse phenol red secretion test;observe asthma incubation period through mouse histamine phosphate-induced asthma test;investigate its anti-inflammatory effect through xylene-induced mouse auricle swelling test.(4)Determine the maximum dosage of Jidou Zhike San by conventional methods.Results:(1)the optimal extraction process of Jidou antitussive granules:reflux extraction with 50%ethanol,the ratio of material to liquid is 1:12,three times of extraction and three hours for each time,combine filtration after filtrate,decompression concentration to obtain dry cream.(2)The shaping process of Jidou antitussive granulesi:mix the dry cream powder and the auxiliary material lactose 4:1,use 50%ethanol as the wetting agent to make soft materials,then pass 14 mesh sieve to make granules,dry at 60? for 1 hour,then screen the whole granules at 10 mesh.(3)The results showed that the cough times of mice in the low,middle and high dose group of Jidou antitussive granules were significantly reduced(P<0.05,P<0.01),and showed a dose-response relationship;the phenol red excretion in the trachea of mice in the high dose group of Jidou Zhike San was significantly increased(P<0.05);the asthma latency of mice in the high dose group in both Jidou antitussive granules and Jidou Zhike San were significantly prolonged(P<0.01);the paw swelling rate of mice in middle and high dose group of Jidou Zhike San was significantly reduced(P<0.05);(4)the results of maximum dose measurement showed that when the dose was 11.7g.kg-1,the mice did not die,and no abnormality was found in liver,kidney,spleen,lung,heart,thymus and other organs of each mouse.Conclusion:(1)the best extraction and preparation process of Jidou antitussive granulesi was optimized through the process of extraction and preparation.(2)It was confirmed that Jidou antitussive granules has good antitussive and antiasthmatic effects through the main pharmacodynamics research.However,Jidou Zhike San has no obvious antitussive effect,and only shows certain expectorant,antiasthmatic and anti-inflammatory effects.Jidou antitussive granules are worthy of further research and utilization.
