Objective: To study the relationship between the expression of rhoguanine nucleotide exchange factor 15(Arhgef15)and its downstream factor RhoA in the brain tissue of patients with intractable epilepsy,and to explore the possible pathway mechanism of epilepsy.Methods: The epilepsy patients who were operated in the Affiliated Hospital of Jining Medical University were selected as the case group and the patients who were operated in the same period as the control group.Changes in the expression of Arhgef15 and RhoA in each group were detected using Western blot and immunohistochemistry.Results: Western blotting showed that there was no expression of arhgef15 in both groups,western blot and immunohistochemical results showed that the protein expression level of RhoA was significantly higher in epileptic patients.Conclusion:(1)Arhgef15 is not expressed in the adult brain.(2)RhoA is differentially expressed in the brain tissue of epileptic patients and normal people.(3)The abnormal expression of RhoA may be a key factor in the epileptogenic pathway in epileptic patients.
