Objective: Alcohol dependence is a serious problem in today's society.It not only causes physical and mental disorders to alcohol dependent individuals,but also causes heavy family and social burdens.In this study,the correlation between GRIK1 gene,KLB gene,AUTS2 gene and GRM8 gene and alcohol dependence was further studied by correlation statistical analysis of genotype and allele frequency of GRIK1 gene,KLB gene,AUTS2 gene and GRM8 gene in alcohol dependence and healthy control group.Methods: By means of cross-sectional investigation,205 male patients with alcohol dependence in line with the international classification of Diseases-10(ICD-10)were selected,and 165 normal controls were selected from the sample bank of Jining Medical College.2.The genotypes and alleles of GIRK1,KLB,aust2,rs9886351,GRM8 and rs886003 were detected by polymerase chain reaction(PCR)Hardy Weinberg genetic balance test was used for this genotype,and chi square test was used for data and allele frequency comparison between different genotypes.The inspection level is ? = 0.05.Results:1.Rs2832407,rs11940694,rs6943555,rs9886351,rs886003 of GIRK1,rs6943555,rs9886351,and rs886003 of GRM8 were in accordance with Hardy Weinberg genetic balance(P = 0.583 and 0.985,P= 0.308 and 0.512,respectively)P = 0.141 and 0.998,P = 0.468 and0.556,P = 0.228 and 0.329,all greater than 0.05),indicating that the samples selected in the experiment are very representative.2.The genotype and allele frequency of rs2832407 at SNP site of grik1 gene were significantly different between alcohol dependent case group and healthy control group(P = 0.026,P = 0.000).C allele might be alcohol dependent susceptibility gene.3.The distribution of genotype and allele frequency of SNP site rs11940694 of KLB gene between alcohol dependent patients and healthy controls was significantly different(P = 0.016,P = 0.031).4.AUTS2 The genotype and allele frequency of SNP rs6943555 were significantly different between alcohol dependent case group and healthy control group(P = 0.042,P = 0.012);the genotype frequency of SNP rs9886351 was significantly different between alcohol dependent case group and healthy control group(P = 0.049),while the allele frequency distribution was significantly different(P = 0.049)There was no statistical significance(P = 0.117).5.The genotype and allele frequency of rs886003 in GRM8 SNP locus were significantly different between alcohol dependent case group and healthy control group(P = 0.003,P = 0.025).Conclusion:1.The SNP of grik1 gene rs2832407 was correlated with alcoholdependence,and C allele might be the susceptible gene of alcohol dependence.2.Alcohol dependence was correlated with KLB rs11940694.3.Alcohol dependence was correlated with AUTS2 rs6943555.The genotype frequency distribution of rs9886351 was statistically significant.The difference of allele frequency distribution was not statistically significant.4.There was a correlation between alcohol dependence and rs886003.
