Objective: Under the guidance of Professor Jiang Lijuan's clinical “Hechang” thought,explore the experience of national famous Chinese medicine professor Zhang Liangying Fang Yangyang Jianpi Recipe(Atractylodes macrocephala 15 g,Chuanxiu Duan 15 g,mulberry parasite 15 g,dodder 20 g,epimedium 15 g,astragalus 20 g,Licorice 6g,Codonopsis pilosula 20 g,Poria 15 g,Eucommia ulmoides 15 g,Ejiao 15g),through the detection of changes in the expression levels of Foxp3 and RORg?t in the decidua tissue of abortion model mice,explore the mechanism of Wenyang Jianpi Recipe in the treatment of unexplained recurrent abortion.Methods: 1.Experimental method: construct CBA/J female mice×DBA/2 pregnant mice as animal models of unexplained recurrent miscarriage diseases,select CBA/J female mice×BALB/c pregnant mice in the normal pregnancy group,and group 10 mice into a group The normal pregnancy group is a blank control group,and the components of the URSA mouse model are the Wenyang Jianpi Recipe group,the dydrogesterone group,and the model control group.The Wenyang Jianpi Recipe group is randomly divided into three dose groups according to the administered dose,namely(Low,medium and high doses).After giving different interventions,mice in each group took their decidual tissues on the 14 th day of pregnancy and conducted the following experiments:(1)Flow cytometry(FACs)was used to detect the Th17 cell-specific transcription factor retinoic acid lone receptor ?t(RORg?t)and Treg cell-specific transcription factor forkhead-like transcription factor p3(Foxp3)expression levels;(2)Counting the embryo loss rate of each group of mice;(3)ANOVA method using multiple samples,using SPSS22.0 The software was used to control the forkhead-like transcription factor p3(Foxp3)and tretinoin in the blank control group,didyrogesterone group,Wenyang Jianpi prescription group(low,medium and high dose)and model control mice.The expression level of orphan receptor ?t(RORg?t)was different,and the embryo loss rate of each group was counted and data analysis was conducted.2.Read ancient classic literature,learn about the etiology,pathogenesis and treatment of slipping tires;check the research and development of unexplained recurrent miscarriage by everyone in modern Chinese medicine schools;by reading Chinese and foreign literature related to unexplained recurrent abortion To further understand the pathogenesis,diagnosis and treatment of unexplained recurrent miscarriage,and to understand the new progress and new directions of the disease.Results: 1.The embryo loss rate of the blank control group was significantly lower than that of the model control group,P<0.01,which was statistically significant;the low-dose group of Wenyang Jianpi Recipe,the middle-dose group of Wenyang Jianpi Recipe,the highdose group of Wenyang Jianpi Recipe,and Diqu pregnancy The rate of embryo loss in the ketone group was significantly lower than that of the model control group,P<0.05,which was statistically significant.There was no statistically significant difference in the rate of embryo loss.2.Compared with the blank control group,the RORg?t level of the model control group was significantly increased(P<0.01),and the Foxp3 level was significantly reduced(P<0.01);the didyrogesterone group,the low-dose group of Wenyang Jianpi Recipe and the middle-dose of Wenyang Jianpi Recipe There were no statistically significant differences in the levels of RORg?t and Foxp3 between the two groups,the high-dose group of Wenyang Jianpi Recipe,and the Foxp3 level;compared with the model control group,the group of didyrogesterone,the low-dose group of Wenyang Jianpi Recipe,the medium-dose group of Wenyang Jianpi Recipe The RORg?t level decreased and the Foxp3 level increased in the high-dose group of Wenyang Jianpi Recipe.Among them,the middle-dose group(P<0.05)and the dydrogesterone group(P<0.05)had statistical differences.Conclusion: 1.CBA/J×DBA/2 mice were used to construct an abortion mouse model.Experiments showed that the model mice in different groups all had a decrease in Foxp3 levels in pregnant decidual tissues and an increase in RORg?t levels.2.Different doses of Wenyang Jianpi Recipe can increase the level of Foxp3 in uterine decidual tissue of URSA model mice in different degrees,and can also reduce the level of RORg?t in different degrees.Compared with the model control group,the middle dose group and the dydrogesterone group are more Significantly,while there was no significant difference between the low,medium and high dose groups.3.As one of the specific transcription molecules of Treg cells,when Treg cells are in a dominant position,Foxp3 and related cytokines are highly expressed in decidual tissues;while as one of the specific transcription molecules of CD4+T cells Th17 cells,when When Th17 cells are in a dominant position,RORg?t and related cytokines are highly expressed in decidual tissues.By examining the expression levels of Foxp3 and RORg?t in pregnant decidual tissues of experimental mice in each group and the embryonic absorption rate of mice in each group,it can be deduced that the upregulation of Foxp3 expression level and the downregulation of RORg?t expression level may be the mechanism by which Wenyang Jianpi Recipe can play a role in preserving the fetus.RORg?t is highly expressed in the model control group(ie,URSA mouse model).After the intervention of Wenyang Jianpi Recipe and Dydrogesterone,this dominant expression has been reversed to a certain extent,showing the dominant expression of Foxp3.Therefore,it can be seen that Wenyang Jianpi Recipe can replace the dominant expression of Foxp3 with the dominant expression of RORg?t,which makes the Th17/Treg ratio in the abortion model return to the normal ratio and restore normal pregnancy.
