Objective: To explore the ability of puerarin combined with adipose-derived stem cells(ADSCs)scaffold to repair tibial defect in postmenopausal osteoporotic(PMOP)rats through the in vivo experiment of Sprague Dawley(SD)rats.Methods:(1)36 3-month-old female SD rats were used to establish models of postmenopausal osteoporosis by bilateral ovariectomy.three months after successful modeling,bilateral proximal tibial bone defects were established,and then were randomly divided into three groups according to different intervention measures: group A,ADSCs-loaded hydroxyapatite stents were implanted in the right tibia defect,and no special treatment was given to the left tibia defect as the control group.Puerarin injection was injected intraperitoneally for 2 g/d.In group B,ADSCs-loaded hydroxyapatite stents were implanted in the right tibial defects,and the left side was not treated as a control,and normal saline was injected intraperitoneally in group C.ordinary hydroxyapatite stents were implanted in the right bone defects in group C,and normal saline was injected intraperitoneally in the left untreated group.The body weight,body fat rate,bone mineral density and pathological sections of the left and right tibia were observed in the three groups at 2,4,8 and 12 weeks after operation,respectively.(2)MEDLINE,Web of Science,EBSCO,The Cochrane Library and other databases were searched by computer,and the related gene targets of puerarin in the treatment of osteoporosis were screened and analyzed by bioinformatics.Results:(1)After 8 weeks,the bone mineral density of group A was higher than that of group B and C,the body weight of group A was lighter than that of group B and C,and the body fat rate of group A was lower than that of group B and C.the general observation of tibial defect showed that there was no significant difference among group A,B and C at 8 weeks,but the repair of bone defect was found in group A > group B > group C after 4 weeks.Pathological sections showed that at the same time,the accumulation,infiltration and new bone formation of inflammatory cells in group A were higher than those in group B and C.(2)Puerarin in the treatment of osteoporosis may be related to prolactin signal pathway,thyroid hormone signal pathway,autophagy regulation,estrogen signal pathway,osteoclast differentiation and endometrial carcinoma related pathway.Conclusions:(1)Puerarin combined with ADSCs containing hydroxyapatite scaffold can effectively promote the healing of tibial defect in postmenopausal osteoporosis rats.Application of puerarin more than 2 weeks can effectively increase bone density,reduce body fat rate and improve postmenopausal osteoporosis.(2)The results of bioinformatics analysis showed that puerarin in the treatment of osteoporosis may be related to prolactin signal pathway,thyroid hormone signal pathway,autophagy regulation,estrogen signal pathway,osteoclast differentiation and endometrial carcinoma related pathway.
