Objective: The effect and possible mechanism of norcantharidin on the proliferation,apoptosis,and invasion of CRC cells.Colon cancer cell lines Caco2 or HT-29 were treated with norcantharidin chewing hormone at a concentration of 0-120 ?M.The cell proliferation rate was determined by CCK-8 method,and apoptosis was detected by Hoechst-33258 staining.Transwell assay was used to assess the cell invasion.Western blot was used to analyze changes in apoptosis and EMT pathway molecules.Methods: CRC cell lines Caco2 or HT-29 were treated with norcantharidin at a concentration of 0-120 ?M.The cell proliferation was determined by the CCK-8,and apoptosis was detected by the Hoechst-33258 staining.Transwell assay was used to evaluate the cell invasion.Western blot was used to analyze changes of apoptosis and EMT pathway molecules in protein levels.Results: Norcantharidin inhibits cell proliferation and invasion,at the same time it promotes apoptosis.Western blot analysis showed that norcantharidin upregulated Bax,cleave caspase-3,and cleave caspase-9in a concentration-dependent manner,and downregulate Bcl-2 to affect the apoptosis of colon cancer cells.Norcantharidin upregulated E-cadherin in a concentration-dependent manner,and downregulated the levels of N-cadherin,Vimentin,p-ERK and integrin ?6 molecules to affect CRC cell invasion.Conclusion: These results indicate that norcantharidin regulates apoptosis through the Bcl-2 signaling pathway and regulates cell invasion by affecting EMT pathway molecules,ERK phosphorylation,and integrin beta6 levels.
