Objective:To detect the expression of Ubiquinol-cytochrome c reductase core protein I(UQCRC1)in human lung adenocarcinoma and to explore the correlation between the expression of UQCRC1 in human lung adenocarcinoma tissue and serum.Methods: 30 fresh human lung adenocarcinoma tissues(excluding other cancerous components)and 30 matched paracancerous fresh lung tissues(5cm away from the edge of the tumor tissue)were collected from patients undergoing thoracic surgery in our hospital in 2017.The cutting edges of all the fresh lung tissues adjacent to the cancer were confirmed to be cancer-free by pathological sections.Another 30 fresh lesions of human lung benign lesions were collected,including pulmonary tuberculosis,pulmonary inflammatory pseudotumor,pulmonary sclerosing hemangioma,pulmonary granulomatous inflammation and so on.The fasting blood samples of all patients were collected 1 day before operation and 7 days after operation.After centrifugation within 2 hours,the serum was collected and stored at-80 ?.Tissue pantothenol-cytochrome c reductase core protein 1 was determined by immunohistochemical method.The concentration ofpantothenol-cytochrome c reductase core protein 1 in peripheral serum was determined by ELISA method.T-test was used for statistical paired samples,and rank sum test was used for immunohistochemistry.Results: Histological results: UQCRC1 was expressed in human lung tissues in different degrees.The average immune score was 8.10 ±2.13 in human adenocarcinoma,6.13 ±3.49 in paracancerous lung tissues and 6.30±3.07 in benign lung lesions(P<0.05).The difference was statistically significant(P < 0.05).The average expression score of UQCRC1 in lymph node metastasis positive group was 9.53 ±1.58,and that in negative group was 7.57 ±2.15(P < 0.05),indicating that the expression of VEGF in lymph node positive group was higher than that in lymph node negative group(P<0.05).ELISA results: the concentrations of UQCRC1 in peripheral blood of patients with lung adenocarcinoma and patients with benign pulmonary diseases on the 7th day after operation were 147.39 ±35.77ng/mL and150.31 ±38.63ng/mL respectively,which were significantly higher than those on the first day before operation(106.15±33.87ng/mL,109.42±40.03ng/mL P=0.000).The difference was statistically significant.One day before operation,the concentration of UQCRC1 in patients with lung adenocarcinoma was 106.15 ±33.87 ng / ml,and that in patients with benign lung diseases was 109.42 ±40.03 ng / ml P=0.734.The concentration of UQCRC1 in patients with lung adenocarcinoma was 147.39 ±35.77 ng / ml at 7 days after operation,and that in patients with benign lung diseases was150.31 ±38.63 ng / ml P=0.762,which was statistically insignificant.Conclusion: 1.The expression of UQCRC1 in human lung adenocarcinoma was higher than that in human benign lung lesions and paracancerous tissues.2.The expression of UQCRC1 in human lung adenocarcinoma with positive lymph node metastasis was higher than that in human lung adenocarcinoma with negative lymph node metastasis.3.The concentration of UQCRC1 in peripheral serum of human lung adenocarcinoma 7 days after operation was higher than that of 1 day before operation,which may be caused by the cleavage or release of mitochondria after operation.
