Objective:To investigate the effects of hyperthermia and PES?2-Phenylethynesulfonamide?on the proliferative activity of gastric cancer SGC7901 cells and the killing ability of NK cells,and intend to provide experimental basis for cellular immunotherapy based on hyperthermia.Methods:Human gastric cancer SGC7901 cells were divided into four groups:37?group,37?PES group,43?group and 43?PES group.The cells in 37?group were cultured at 37?5%CO2 for 1 hour,and the cells in 43?group were cultured at 43?5%CO2 for 1 hour.PES group was added with 15 umol/L PES at different temperatures.The cells of each group were cultured in 37?5%CO2 medium for 6 hours after preliminary culture under set conditions,and then NK cells were co-cultured for 24 hours in each group.CCK-8 assay was used to detect the effect of hyperthermia and PES on proliferation of gastric cancer SGC7901 cells,so as to the killing rate of NK cells on gastric cancer SGC7901 cells.ELISA assays was used to detect the release of perforin and granzyme B levels of NK cells in each group.Results:1.The results of CCK-8 assay showed that the inhibition rates of gastric cancer SGC7901 cells treated with hyperthermia in three different time periods?6h,12h,24h?were16.65±3.66,8.68±2.53,32.94±1.66respectively,the proliferation inhibition rates of PES on gastric cancer SGC7901 cells were 26.77±2.930,22.14±5.51,40.69±1.79,respec-tively,the inhibition rates of hyperthermia and PES on the proliferation of gastric cancer SGC7901 cells were 41.81±3.63,34.80±5.27,47.59±1.60,respectively.The cell inhibition rate in 43?PES group was highest than37?PES group and 43?group,37?PES group was higher than 43?group?P<0.05?.The killing rate of NK cells in each group at 6h was56.37±2.21,7.34±4.14,64.91±5.78,4.04±2.14 in 37?group,37?PES group,43?group and 43?PES group;In these groups,at 12h,the killing rates of NK cells were 64.16±3.37,11.59±6.15,74.22±0.51 and28.31±4.42,respectively.The killing rate of NK cells in each group at24h was 85.40±2.26,35.37±2.37,90.41±2.79 and 57.12±4.11.The killing rate of NK cells in 43?group was more highest than 37?group,43?PES group and 37?PES group,which 37?group was highest than43?PES group and 37?PES group,and 43?PES group was higher than37?PES group?P<0.05?.2.The results of ELISA assay:the granzyme B level of NK cells at6h after adding NK cells?pg/ml?was 2820.61±8.35,2240.77±5.43,3145.24±15.33 and 2469.35±30.67 in each group,respectively;detection of perforin?pg/ml?was 1661.65±9.34,1054.58±14.24,2072.03±31.09,1146.64±11.76 in each group,respectively.The granzyme B?pg/ml?of NK cells in each group at 12h was 3310.45±11.15,2735.19±17.20,3673.27±9.26 and 3038.14±28.43 in each group,respectively;detection of perforin?pg/ml?was 2213.79±34.33,1311.89±29.72,2311.69±28.41and 1920.54±29.23 in each group,respectively.At 24 h,the granzyme B of NK cells in each group?pg/ml?was 3710.52±7.38,3037.29±11.54,3928.80±25.75 and 3559.45±39.12 in each group,respectively;detection of perforin?pg/ml?:was 2281.08±35.37,1879.90±29.71,2476.45±13.57and 2187.98±35.04 in each group,respectively.The comparison of granzyme B and perforin concentration released by NK cells in 43?group was more highest than 37?group,43?PES group and 37?PES group,which 37?group was highest than 43?PES group and 37?PES group,and 43?PES group was higher than 37?PES group?P<0.05?.Conclusions:1.Hyperthermia can enhance the inhibitory effect of PES on the proliferation of gastric cancer SGC7901 cells;2.Hyperthermia can enhance the killing effect of NK cells on gastric cancer SGC7901 cells,but PES can inhibit the killing ability of NK cells.
