Objective:In this study,plasma transcriptome microRNA?miRNA?was sequenced in 3 high-risk people with ischemic stroke without ischemic stroke and 24 hours after the onset of ischemic stroke.53 miRNAs with differential expression were screened out miR-423-5p is the most significant miRNA.Real-time fluorescence quantitative PCR to further verify the expression level of miR-423-5p in healthy people,high-risk ischemic stroke population and patients with ischemic stroke within 24h,to evaluate miR-423-5p in ischemic stroke The potential clinical significance of early warning and diagnosis.Methods:The peripheral venous blood samples of 3 high-risk populations with ischemic stroke and no-ischemic stroke and their onset were collected for high-throughput sequencing of transcriptomics to obtain differential miRNA?miR-423-5p?.Venous blood samples were collected from46 patients with ischemic stroke within 24 hours,43 patients with high risk of ischemic stroke and 46 healthy people.The plasma was separated by centrifugation,and then the total RNA in the plasma was extracted and reverse transcribed into cDNA.The real-time fluorescence quantitative PCR method was used to detect the expression of miR-423-5p in the three plasmas,and U6was used as an internal reference.Using 2-??Ctalgorithm to calculate and obtain the relative expression of miRNA?miR-423-5p?,statistical analysis of miR-423-5p in healthy people,high-risk population of ischemic stroke and stroke onset 24h three Different groups of plasma expression differences.Results:1?Through high-throughput sequencing of miRNA,a total of 53differentially expressed miRNAs were screened from the 3 patients before and after the onset of ischemic stroke,of which miR-423-5p was the one of the most significant microRNA.2?The expression of plasma miR-423-5p in the ischemic stroke high-risk group was higher than that in the ischemic stroke group and the healthy control group?p<0.001?;the plasma miR-423-5p in the ischemic stroke group423-5p was up-regulated relative to the healthy control group?p<0.001?.3?The ROC curve shows that miR-423-5p is of moderate or higher diagnostic value for early warning of ischemic stroke.Conclusions:1?High-throughput sequencing results found that the expression of miR-423-5p was relatively down-regulated after ischemic stroke in high-risk people with ischemic stroke,suggesting that it may be involved in the occurrence and development of ischemic stroke.Of biomolecular markers for early warning and early diagnosis of sexual stroke.2?miR-423-5p is up-regulated in plasma of patients with ischemic stroke compared with healthy people;compared with high-risk population of ischemic stroke,miR-423-5p is down-regulated in patients with ischemic stroke miR-423-5p has a high diagnostic value for early warning of ischemic stroke,and the value of early diagnosis of ischemic stroke still needs further discussion.
