Objective The purpose of this study is to explored the correlation between serum levels and insulin sensitivity in newly diagnosed type 2 diabetes mellitus patients.Methods In this study,a total of 66 newly diagnosed type 2 diabetes mellitus patients and 40 impaired glucose tolerance patients were recruited from the outpatient department of the first affiliated hospital of the university of south China from October 2018 to April 2019,and 43 normal glucose tolerance from the physical examination center during the same period were used as the control group.Basic data of all subjects were collected,including age,gender,past history,family history,personal history(alcohol and tobacco history)and recent medication status.Height,weight,waist circumference,blood pressure,fasting glucose,OGTT 30 min glucose,OGTT 60 min glucose,OGTT 120 min glucose,fasting insulin,OGTT 30 min insulin,OGTT 60 min insulin,OGTT120 min insulin,total cholesterol,triglyceride low-density lipoprotein cholesterol.high-density lipoprotein cholesterol,glycosylated hemoglobin,creatinine,uric acid,urea nitrogen,alanine aminotransferase,glutamate aminotransferase,high-sensitivity c-reactive protein were determined for all subjects.Fasting serum levels of MOTS-c in all subjects were measured by enzyme-linked immunosorbent assay.Body mass index,body fat fraction,area under insulin curve,area under glucose curve,insulin resistance indexhomeostasis model evaluation and insulin secretion homeostasis model evaluation were calculated.In addition,25 healthy subjects were randomly selected,the level of their serum MOTS-c was measured at various time points during OGTT.Results Serum MOTS-c increased gradually in NGT,IGT and T2DM(12.88?11.44 vs 39.56?22.87 vs 72.36?19.78 ng/mL,P < 0.001).In all subjects,serum MOTS-c was positively correlated with age,BMI,waist circumference,BF%,FPG,AUC-glucose,HOMA-IR,and negatively correlated with HOMA-?(P < 0.05).Multiple linear regression showed that age,BMI and AUC-glucose were independent correlation factors of serum MOTS-c.Multivariate logistic regression analysis confirmed that MOTS-c was independently and positively correlated with IR and T2 DM.MOTS-c predicted that the area under the ROC curve of T2 DM was 0.785(95% CI:0.715-0.842).The OGTT test showed that the fluctuation curve of serum MOTS-c was similar to that of glucose and insulin,and the area under the curve was significantly positively correlated with each other(P < 0.001).Conclusion The expression of serum MOTS-c was significantly increased in newly diagnosed type 2 diabetes mellitus patients,which was related to insulin sensitivity.MOTS-c may be involved in the regulation of glucose metabolism.
