Objective and Background: Prior research has found that DACT3 may have tumor-suppressing effects in tumorigenesis.but its role in papillary thyroid carcinoma is still unclear.This study attempts to compare the expression of DACT3 in papillary thyroid carcinoma and in adjacent nonneoplastic tissue,and to identify potential correlation of the expression profile with clinicopathology.The author aims to provide new insights into its role in the diagnosis and risk assessment,and as a therapeutic target,of papillary thyroid carcinoma.Methods: 1.Study target DACT3 was selected by screening the following databases: KEGG,TCGA and GEPIA.Its expression data in thyroid cancer and adjacent normal tissue were obtained and potential correlation of DACT3 expression level with thyroid cancer clinical behavior was analyzed in terms of survival and risk stratification.2.Fresh tumor tissue and adjacent non-neoplastic tissue were collected intraoperatively from forty-eight(48)newly diagnosed papillary thyroid carcinomas.RNAs from these paired samples were extracted and used for measuring DACT3 expression by qRT-PCR;formalin-fixed and paraffin embedded tumor and non-neoplastic tissues were then used for the detection of DACT3 protein expression by immunohistochemistry.3.Statistical analysis was performed to explore the relationship between DACT3 expression and clinicopathologic characteristics in thyroid papillary carcinoma.Results:1.Bioinformatics analysis revealed that the expression of DACT3 in thyroid cancer tissue was significantly lower than the adjacent non-neoplastic counterpart;the expression of DACT3 was negatively correlated with the survival rate and survival time of thyroid cancer;the expression of DACT3 was positively correlated with the risk classification of thyroid cancer.2.qRT-PCR mRNA level analysis showed that the expression of DACT3 in human thyroid papillary carcinoma tissues was significantly lower than adjacent non-neoplastic tissues,and the expression of DACT3 was related to tumor size.3.Immunohistochemical results demonstrated that the positive staining rate of DACT3 in papillary thyroid carcinoma tissues was significantly lower than that in adjacent tissues,and its expression did not show any significant relationship with patient gender,age,tumor size,cervical lymphatic metastasis,TNM staging,distant metastasis,risk degree,or Ki-67 labeling index.Conclusion: The low expression of DACT3 in clinical tissue samples of human thyroid papillary carcinoma was significantly related to tumor size of thyroid papillary carcinoma;the positive staining rate of DACT3 in papillary thyroid carcinoma is lower than adjacent tissue.
