Objective:To study the effect of PES on Fascin expression,migration and invasion of BGC-823 cells,which provides a theoretical basis for the clinical treatment of PES in gastric cancer.Methods:The poorly differentiated human gastric cancer BGC-823cell line were cultured in 37℃,5%CO2 cell incubator in vitro to grow the cells to the logarithmic growth phase,and the gastric cancer BGC-823cells were divided into three groups:PES group,IMD-0354(NF-κB signaling pathway inhibitor)group and control group.The above grouped cells were treated separately according to the conditions set in the experimental grouping,and culture was continued for 48 hours.Then conduct the following experiments:1.the expression of Fascin protein and NF-κB p65 in gastric cancer BGC-823 cells were detected by Western blot;2.Transwell assays were used to detect the migration and invasion number of gastric cancer BGC-823 cells.Results:1.Western blot analysis for the expression of Fascin protein and NF-κB p65 in gastric cancer BGC-823 cells:(1)The gray values of Fascin protein expression of gastric cancer BGC-823 cells in different groups:PES group 5.63±0.80,IMD-0354 group 12.02±0.70,control group 44.98±0.69,and the difference analysis of the expression of Fascin protein of gastric cancer BGC-823 cells in control group was highest than IMD-0354 group and PES group(P<0.05).(2)The gray values of NF-κB p65 expression in gastric cancer BGC-823 cells under different grouping conditions were:PES group 5.56±0.72,IMD-0354 group 18.02±0.66,control group 39.86±0.78,and the difference analysis of the expression of NF-κB p65 in gastric cancer BGC-823 cells compared in control group was highest than IMD-0354 group and PES group(P<0.05).2.The number of migration and invasion of gastric cancer BGC-823cells in each group was detected by Transwell chamber:(1)The migration number of gastric cancer BGC-823 cells:PES group90.33±5.51,IMD-0354 group 115.67±5.13,control group 245.67±6.03,and the migration number of gastric cancer BGC-823 cells after 48h in control group was highest than IMD-0354 group and PES group(P<0.05).(2)Invasion number of gastric cancer BGC-823 cells:PES group 38.00±3.61,IMD-0354 group 63.33±4.16,control group 92.67±3.06,and each group of gastric cancer BGC-823 cells Comparison of the number of invasions after 48h in control group was highest than IMD-0354 group and PES group(P<0.05).Conclusion:1.PES can inhibit the migration and invasion of gastric cancer BGC-823 cells and the expression of Fascin and NF-κB p65;2.PES inhibits gastric cancer BGC-823 cell migration and invasion and Fascin expression.The possible mechanism is related to the activity of NF-κB signaling pathway in BGC-823 cells.
