Objective:The purpose of this study was to investigate whether the single or combined action of the two drugs could change the expression state of the Reprimo gene in human gastric adenocarcinoma cells by using the effect of dicitabine combined with apatinib on the expression of the Reprimo gene,so as to provide experimental basis for the further study of the Reprimo gene.Methods : 1.After being cultured well,gastric adenocarcinoma SNU-1 cell lines were randomly divided into normal saline group,dexitabine group,apatinib group and combination group.After being mixed with CCK-8 reagent and treated with drugs for 24 hours,the morphological changes of gastric cancer cells were observed by light microscope,and the absorbance of each group at 450 nm was measured.2.Real-time PCR(RT-PCR)was used to detect the mRNA expression of reprimo gene in SNU-1 cell lines treated with drugs alone or in combination.3.Western Blotting method was used to analyze the changes of reprimo protein expression in each group of cells after intervention.Results:Compared with the control group,the inhibitory effect of dexitabine combined with apatinib on gastric cancer cell line SNU-1 was the most obvious,and the difference was statistically significant(P< 0.05).When the expression of mRNA was detected by RT-PCT,the expression level of mRNA in the dexitabine group was significantly higher than that in the control group,followed by the combined action of the two groups.(P < 0.05).Western Blotting assay showed that the expression of Reprimo protein increased in different degrees after treatment with dexitabine alone,apatinib group and combination group,and the effect of combination group was the most obvious(P < 0.05).Conclusions : 1.Dexitabine combined with apatinib can better inhibit the proliferation of human gastric adenocarcinoma SNU-1 cells.2.Both dexitabine and apatinib can restore the expression of Reprimo protein in human gastric adenocarcinoma cell line SNU-1 to some extent,and the effect of the combination of the two drugs is better than that of single drug.
