MiR-23c Inhibits Migration And Invasion Of Glioma U87 Cells By Decreasing Mtdh Expression | | Posted on:2020-10-17 | Degree:Master | Type:Thesis | | Country:China | Candidate:J C Zhang | Full Text:PDF | | GTID:2404330602988006 | Subject:Surgery (neurosurgery) | | Abstract/Summary: | | | Objective: To investigate the effect of miR-23 c on proliferation,migration and invasion of the U87 glioma cells.Methods: U87 cells were transfected miR-23 c mimic or miR-NC by liposome,and the miR-23 c expression levels were examined by qRT-PCR.The effects of miR-23 c on cells proliferation,migration and invasion were observed by the MTT assay,cell scratch and Transwell invasion.Bioinformatics analysis revealed that metadherin(MTDH)might be a potential target gene for miR-23 c.Subsequently,Western blot,RT-PCR and luciferase reporter gene were used to detect the effects of miR-23 c on MTDH.Co-transfected with MTDH and miR-23 c to clarify the role of miR-23 c on glioma cells proliferation,migration and invasion by regulat ing MTDH expressi on.Results: MiR-23 c expression was significantly up-regulated in the miR-23 c transfected cells.Mir-23 c significantly decreased the cell proliferation,migration and cell invasion ability.Luciferase reporter gene assay showed that miR-23 c can target bind with MTDH.Furthermore,qRT-PCR and Western bloting suggested that miR-23 c decreased the mRNA and protein expression of MTDH.Compared with the miR-23 c group,the proliferation rate,invasive ability and migration ability of the cells in the miR-23 c and MTDH co-transfected groups were significantly increased.Conclusions: In U87 glioma cells,miR-23 c inhibited the cells proliferation,migration and invasion by targeting and decreasing MTDH expression. | | Keywords/Search Tags: | miR-23c, Glioma, MTDH, proliferation, Invasion, Migration | | Related items |
