Objective: By studying the effect of acupuncture on myocardial fibrosis in rat model of chronic heart failure,the possible mechanism of acupuncture in the treatment of chronic heart failure(CHF)was discussed,providing a basis for further clinical treatment of chronic heart failure by acupuncture.Methods:According to the random number table,32 healthy male clean grade SD rats were divided into normal control group(8),CHF model group(8),sham acupuncture treatment group(8)and acupuncture treatment group(8).Normal control group and CHF model group were not treated.The sham acupuncture group was treated with sham acupuncture for 8 weeks.Acupuncture treatment group was given acupuncture treatment for 8 weeks.The physiological status,B type natfiuretic peptide(BNP),angiotensin II(Ang?),echocardiographic indicators,and myocardial pathological morphology and myocardial fibrosis-related proteins of each group of experimental rats were evaluated before and after the experiment.Before and after intervention,left ventricular end systolic diameter(LVIDs),left ventricular end diastolic diameter(LVIDd),left ventricular fraction shortening(LVFS)and left ventricular ejection fraction(LVEF)were measured by echocardiography.After the intervention,the pathological changes of myocardium were observed by light microscopy,and collagen III and other related proteins were detected by western-bloting.Results:(1)After modeling,CHF model group,sham acupuncture treatment group and acupuncture treatment group experimental rats weight lighter than normal control group(P < 0.05),LVEF,LVFS lower than normal control group(P < 0.05),and LVIDd,LVIDs,BNP,Ang? increased than the normal control group(P< 0.05),the difference was statistically significant,suggesting building success.(2)At the end of treatment,there was no significant difference in the final weight and LVIDd indexes between the acupuncture treatment group and the normal control group.Comparison between the groups showed no significant differences in the final body weight,LVIDd,LVIDs,LVFS and LVEF between the CHF model group and the sham acupuncture treatment group.However,the final body weight of the CHF model group and the sham acupuncture treatment group was less than that of the normal control group and the acupuncture treatment group(P<0.05),and LVIDd was more than that of the normal control group and the acupuncture treatment group.LVIDs in the CHF model group,the sham acupuncture treatment group and the acupuncture treatment group increased compared with the normal control group(P<0.05).Pair-wise comparison between the three groups showed that LVIDs in the CHF model group and the sham acupuncture treatment group increased compared with the acupuncture treatment group(P<0.05).CHF model group,sham acupuncture treatment group and acupuncture treatment group BNP,Ang ? increased compared with normal control group.Pair-wise comparison between the three groups showed that BNP,Ang?in the CHF model group and the sham acupuncture treatment group increased compared with the acupuncture treatment group(P<0.05).LVEF?LVFS in the CHF model group,the sham acupuncture treatment group and the acupuncture treatment group decreased compared with the normal control group(P<0.05).Pair-wise comparison between the three groups showed that LVEF?LVFS in the CHF model group and the sham acupuncture treatment group decreased compared with the acupuncture treatment group(P<0.05).(3)After the treatment,HE staining pathological sections of rat myocardial tissue in each group were observed under light microscope,indicating that no obvious pathological changes were observed in the pathological sections of rat myocardial tissue in the normal control group.In the CHF model group,the sham acupuncture group and the acupuncture treatment group,the experimental rat myocardial tissue showed myocardial cell disorder,hypertrophy or atrophy,edema,nuclear condensation and lysis,cicatronic degeneration,fibrous tissue hyperplasia,and inflammatory cell infiltration,and other changes of varying degrees of myocardial injury.The degree of myocardial injury in the acupuncture treatment group was less than that in the CHF model group and the sham acupuncture treatment group.(4)At the end of treatment,there was no significant difference in the relative expression of GSK-3?,and no significant difference in the indicators in CHF model group and sham acupuncture treatment group.CHF model group,sham acupuncture treatment group and acupuncture treatment group Collagen ?,?-SMA,?-catenin,p-GSK-3 ? relative expression increased compared with normal control group.Pair-wise comparison between the three groups showed that Collagen ?,?-SMA,?-catenin,p-GSK-3?relative expression in the CHF model group and the sham acupuncture treatment group increased compared with normal control group(P<0.05).Conclusion:(1)Acupuncture can reduce chronic heart failure rats plasma BNP,Ang?level.(2)Acupuncture can reduce LVIDd and LVIDs in rat models of chronic heart failure and inhibit ventricular remodeling.(3)Acupuncture can increase LVFS and LVEF in rat model of chronic heart failure and improve cardiac function.(3)Acupuncture can inhibit myocardial fibrosis by reducing the Collagen ?,?-SMA,?-catenin,p-GSK-3 ? relative expression of chronic heart failure rats,its mechanism may be related to the inhibition of Wnt/?-catenin pathway.
