Objective:To explore the expression level of BRD4 in bladder cancer and its effect on the cycle,apoptosis and proliferation of bladder cancer cell line T24.In addition,we further investigated whether gemcitabine could interfere with the expression of BRD4 and thus affect the cycle,apoptosis and proliferation of T24.Methods : Expression levels of BRD4 gene and protein were detected by real-time quantitative PCR(RT-PCR),western blot and immunohistochemistry.Cell cycle and apoptosis of bladder cancer cell lines were detected by flow cytometry,while cell proliferation was detected by cck-8,plate cloning and Ed U.Results:Compared with normal bladder tissues,the expression levels of BRD4 gene and protein were generally up-regulated in bladder cancer tissues(p<0.05).Compared with SV-HUC-1,BRD4 gene and protein were highly expressed in bladder cancer cell line T24(p<0.05).By immunohistochemical found BRD4 most showed a trend of increase in bladder cancer tissue type low BRD4 expression can make the cell cycle arrest in T24 cells in G0 / G1 phase and cell apoptosis rate increased,cell proliferation is restrained gemcitabine can make the T24 cells cut BRD4 gene and protein expression(p < 0.05),and is T24 cell apoptosis rate increased,proliferation significantly suppressed,the cell cycle to G0 / G1 phase increased.Conclusion:BRD4 is highly expressed in bladder cancer.Knockdown of BRD4 can affect T24 cell cycle,apoptosis and proliferation.The effects of gemcitabine on cell cycle,apoptosis and proliferation may be related to the interference with BRD4 expression.
