ObjectiveTo analyze pathogenesis of postmenopausal osteoporosis from a theoretical point of view,and observe the changes of lipid,bone metabolism and Leptin/ ? 2-AR signal pathway of castrated osteoporosis rats through experimental studies.To observe the rapeutical effect and mechanism of Bushen Huatan Recipe on postmenopausal osteoporosis from the angle of adjusting bone metabolism of rats,and provide a scientific basis for the clinical application of compound recipt.Methods1 Theoretical researchThrough systematic review and summarization of ancient and modern Chinese and foreign literature,in order to understand Chinese medicine's understanding of the etiology and pathogenesis of PMOP and its treatment methods,it provides theoretical basis for experimental research.2 experimental researchSPF 6-month-old SD female non-pregnant rats were randomly divided into a sham operation group,a model group,a Levial group,an atorvastatin calcium tablet group,and a Bushen Huatan Recipe group.A rat model of postmenopausal osteoporosis was established by bilateral ovariectomy,and it was administered by gavage at the5 th week after operation.Levial group and atorvastatin calcium tablet group were given 0.23 mg / kg·b·w and 0.92 mg / kg·b ·w,and traditional Chinese medicine Bushen Huatan Recipe 9.4g / kg·b·w gavage.The sham operation group and model group were given the same volume of normal saline.They were administered once a day.After 8 weeks of continuous administration,the rats were anesthetized and sacrificed.The right ventricle blood was centrifuged.Leptin,E2,PINP,?-CTx levels in rat serum,biochemical analyzer to detect TC,TG,HDL-C,LDL-C levels;right femurs of rats were taken,and bone density of femurs of rats was detected by Mico CT;immunity Histochemical method was used to detect the expression of Leptin and ?2-AR protein;PCR method was used to detect the relative expression of Leptin m RNA and ?2-AR m RNA.SPSS 23.0 was used for statistical analysis of variance.The experimental results were expressed as mean ± standard deviation(x ± S).Pairwise comparison between groups was performed by S-N-K test.P<0.05 indicated that the difference was statistically significant.Conclusions1 Theoretical researchReviewing the relevant literature on PMOP from ancient to modern times,the understanding of the etiology and pathogenesis of PMOP mainly includes deficiency of kidney essence,weakness of spleen and stomach,lack of liver blood,and blood stasis.Based on the research on the correlation between PMOP and lipid metabolism disorders,the professor proposed to the Tan Zhuo that phlegm turbidity is also an important factor in the pathogenesis of OP.On the basis of tonifying the kidney,the OP treats OP from phlegm,formulates a new method of replenishing kidney and resolving sputum,and formulates the formula for replenishing kidney and resolving phlegm.This recipe is the clinical experience of his supervisor,Professor Xiang Nan,and has been shown to have significant effects on PMOP in clinical and animal experiments.Whether Bushen Huatan Recipe can affect the expression of Leptin/?2-AR remains to be further studied.2 Experimental researchBone mineral density measured by Micro CT showed that compared with the sham operation group,the bone mineral density of the model group was significantly reduced(P<0.05);compared with the model group,the bone mineral density of the Liweiai group,atorvastatin calcium tablet group,and Bushen Huatan recipe group's bone mineral density was significantly increased(P<0.05),indicating that Bushen Huatan Recipe,Levial and Atorvastatin Calcium Tablets can increase bone density in castrated osteoporosis rats,but the difference between groups was not statistically significant(P>0.05).The results of the automatic biochemical test showed that compared with the sham operation group,the levels of TC,TG,LDL-C and HDL-C in the model group,Levial group,atorvastatin calcium tablet group,and Bushen Huatan Recipe group were increased.The levels decreased(P<0.05);compared with the model group,the levels of TC,TG,LDL-C decreased,and HDL-C levels increased in the Liweiai group,atorvastatin calcium tablet group,and Bushen Huatan Recipe group(P<0.05);Compared with the atorvastatin calcium tablet group,the Bushen Huatan Recipe group has a lowerlevel of regulating lipid metabolism than the atorvastatin calcium tablet group(P>0.05).The ELISA test results showed that compared with the sham operation group,the levels of Leptin and E2 in the model group,Levial group,atorvastatin calcium tablet group,and Bushen Huatan Recipe group were reduced(P<0.05);compared with the model group,By comparison,the levels of Leptin and E2 in the Levial group,atorvastatin calcium tablet group,and Bushen Huatan Recipe group were increased(P<0.05);and the Bushen Huatan Recipe group increased estrogen levels higher than those in Levial group and Atorvastatin calcium tablets group(P>0.05).Serum bone metabolism indicators showed that compared with the sham operation group,the expression levels of P?NP and ?-CTx in the model group,Levial group,atorvastatin calcium tablet group,and Bushen Huatan Recipe group increased(P<0.05);and Compared with the model group,the expression levels of P ? NP and ?-CTx in the Levial group,atorvastatin calcium tablet group,and Bushen Huatan Recipe group were decreased(P<0.05).Atorvastatin calcium tablets group,but there was no statistical significance between the three(P>0.05).Immunohistochemical results showed that the positive expression of Leptin and ?2-AR was brown particles,which were mainly located in the cytoplasm of bone marrow cavity and around osteoblasts.Pathological microscope observation showed that the expression of Leptin in the sham operation group was strongly positive,and the expression in the Bushen Huatan Recipe group,Levial group,Atorvastatin calcium tablets group was positive,and the model group was slightly stained;?2-AR expression in the model group was strongly positive The expressions of Bushen Huatan Recipegroup,Levial group and Atorvastatin calcium group were positive,while the expression of sham operation group was weak.The PCR method showed that compared with the sham operation group,the relative expression of Leptin m RNA in the model group,Levial group,atorvastatin calcium tablets group,and Bushen Huatan Recipe group decreased,and the relative expression of ?2-AR m RNA increased(P<0.05);Compared with the model group,the relative expression of Leptin m RNA in rats in the Levial group,atorvastatin calcium tablets group,and Bushen Huatan Recipe group was significantly increased,and the relative expression of ?2-AR m RNA was significantly reduced(P<0.05);and the relative expression of Leptin m RNA in the Bushen Huatan Recipe group was higher than that in the atorvastatin group(P>0.05).ConclusionBushen Huatan Recipe can increase bone density and estrogen levels in ovariectomized rats,reduce serum bone turnover related indicators,regulate lipid metabolism,increase serum and bone tissue Leptin content,and reduce bone tissue ?2-AR content.The therapeutic effect of formula on castrated osteoporosis rats may be achieved by affecting the expression of Leptin / ? 2-AR,regulating the processes of lipid metabolism and bone metabolism,promoting bone formation and inhibiting bone resorption.
