Objective:The protective effect and mechanism of primary Dendrobium officinale extract on gastric mucosal damage caused by aspirin,and the traditional Chinese medicine fermentation technology has been used to develop fermented beverages of Dendrobium officinale with antioxidant function on the basis of verifying its beneficial effects on stomach.Methods:1.Protective effect of Dendrobium officinale extract on gastric mucosal inflammation in rats:a model of gastric mucosal injury was established by aspirin administration at 300 mg/kg for 5 hours.The hematoxylin-eosin staining method was used to observe the pathological changes of gastric mucosa in each group of rats after modeling.At the same time,the serum and tissues of rats were measured by enzymelinked immunosorbent assay,quantitative real-time PCR,western blot and immunohistochemistry changes in inflammatory factor levels were detected.2.Protective mechanism of Dendrobium officinale extract on gastric mucosal injury:the changes of TFF2,EGF,and EGFR levels in serum and tissues of each group were detected by enzyme-linked immunosorbent assay,quantitative real-time PCR method,western blot method,and immunohistochemical method.3.Study on the beneficial effects of fermented material of Dendrobium officinale on the stomach and the development of fine products:orthogonal test was used to select the optimal fermentation process.Macroscopic observation of the pathological changes of Dendrobium officinale on rat gastric mucosa,detection of changes in total anti-oxidant capacity before and after fermentation,andobservation of changes in the appearance and taste of beverages after fermentation.Results:1.The extraction rate of Dendrobium officinale extract is 18.2%,polysaccharide content is 67.18%,and protein content is 12.38%.2.Evaluation of rat acute gastric injury model and improvement of pathological injury:Dendrobium officinale extract can significantly improve the gastric mucosal redness,bleeding,erosion caused by aspirin,and it is positively correlated with the dose.3.Changes in the expression of inflammatory factors and growth regulators:preparation of Dendrobium officinale extract can significantly improve the excessively high levels of inflammatory factors and growth regulators when gastric mucosal damage occurs,and its efficacy is positively correlated with dose.4.Effect of fermentation on pharmacological effects of Dendrobium officinale:after fermentation under optimal conditions,the total antioxidant capacity was 2.77 times that before fermentation,and the efficacy,appearance,and taste of the fermented material in improving gastric mucosal damage were significantly better than the extract.Conclusion:1.Dendrobium officinale extract can significantly improve the acute gastric mucosal injury in rats caused by aspirin.2.The protective mechanism of Dendrobium officinale extract may be related to reducing the expression of inflammatory factors and activating the TFF2 / EGF / EGF R pathway to repair the damaged gastric mucosa in time.3.Fermentation can enhance the protective effect of Dendrobium officinale on the gastric mucosa and its antioxidant capacity,and can make Dendrobium officinale look and taste better than before.
