Objective: To study the effect of emodin combined with cisplatin on the invasion and migration of liver cancer HepG2 cells,and to explore the synergistic effect of emodin and cisplatin on liver cancer.Methods: Live cancer HepG2 cells were routinely recovered and cultured in vitro,and cells in the logarithmic growth phase were taken for experiments.HepG2 cells were intervened with different concentrations of emodin and cisplatin alone or in combination with effective concentrations(that is,divided into blank control group,emodin group,cisplatin group and emodin combined with cisplatin group).The cell proliferation rate of cells in each group was detected by MTT reduction method after 24 hours.The cell migration rate of cells in each group was measured by scratch test at 0,12 and 24 hours.Using high content cell analysis technology to observe cell invasion and migration.The expression of E-cadherin and Vimentin in the drug cells of each group was detected by immunofluorescence.Results: 1.The results of MTT assay suggest that compared with the blank control group,HepG2 cells treated with emodin or cisplatin alone show obvious growth inhibition and concentration-dependent,while the combined application of emodin and cisplatin can enhance Compared with other groups,this inhibitory effect is statistically significant.2.The results of the scratch test showed that the wound healing rate was significantly lower than that of the cisplatin or emodin group alone after 0,12,24 hours of combined effect of emodin and cisplatin,and it was concentration and time dependent.The high-content cell analysis experiment showed that the cell migration speed and distance of the combined application group of emodin and cisplatin were significantly reduced compared with the single cisplatin and emodin.3.Immunofluorescence results showed that compared with the single cisplatin and emodin,the expression of E-cadherin in cells of the combined application group of emodin and cisplatin was significantly increased,but Vimentin had no significant changes.Conclusion: The combined application of emodin and cisplatin can inhibit the proliferation,invasion and migration of liver cancer cells;emodin can increase the sensitivity of cisplatin to the above effects,and the two have a synergistic effect.
