Objective:To investigate the effect of Yiqi Jianpi Huashi Recipe on the efficacy of mice with low gastrointestinal motility due to spleen deficiency and its mechanism in the signaling pathway of anti-oxidative stress,and to provide clinical use of Yiqi Jianpi Huashi Recipe to treat gastrointestinal motility experimental basis.Methods:60 mice were taken,10 in the normal group and 10 in the model group,20 in the mosapride group and the Yiqi Jianpi Huashi Recipe group.Except for the normal group,the mice in each group were prepared with rhubarb decoction and gavage into the pathological model of spleen deficiency.After successful modeling,the mice in the mosapride group were given 2.5mg·kg-1of mosapride suspension,Yiqi Jianpi Huashi Recipe group were given 36.8g·kg-11 Yiqi Jianpi Huashi decoction,and the normal and model groups were given equal volumes of distilled water,7 consecutive days of treatment.After treatment,10 mice in each of the mosapride group and the Yiqi Jianpi Huashi Recipe group were sacrificed by neck removal.After feeding for 3 days,the other 10 mice in the normal group,the model group,the mosapride group,and the Yiqi Jianpi Huashi Recipe group were sacrificed by neck removal.Testing relative retention rate of the pigment inside the stomach and small intestine propulsion in mice rate,Hematoxylin-eosin?HE?staining was used to observe the gastric mucosa and duodenal mucosa pathology,protein imprinting method?Western blot?in detecting duodenal tissue in mice Kelch-like ECH-related protein 1?Keap1?,nuclear factor erythroid 2 related factor 2?Nrf2?,and heme oxygenase-1?HO-1?protein expression.Results:Compared with the normal group,the relative residual rate of gastric pigment in the model group was significantly increased?P<0.01?,and the small intestine propulsion rate was significantly reduced?P<0.01?.HE staining display,edema of gastric mucosa in model mice,duodenal villi were broken.Western blot results show the expression of Keap1in duodenal tissue was significantly increased?P<0.01?,the expressions of Nrf2 and HO-1were significantly reduced?P<0.01?.Compared with the model group,the relative residual rates of gastric pigment in the mice in each treatment group were significantly reduced?P<0.01?.The small intestine propulsion rate of mice in mosapride group and Yiqi Jianpi Huashi Recipe group increased significantly?P<0.01?.3 days after the end of dosing,compared with the model group,the small intestine propulsion rate of mice in Yiqi Jianpi Huashi Recipe group still increased significantly?P<0.01?,but there was no significant difference in small intestinal propulsion rate in mice of mosapride group.Gastric mucosal cells and duodenal villi of mice in the treatment group were repaired to varying degrees.The expression of Keap1 in the duodenal tissues of the treatment groups was significantly reduced?P<0.01?,and the expressions of Nrf2 and HO-1 were significant increased?P<0.01?.Conclusion:Yiqi Jianpi Huashi Recipe can fundamentally improve the low gastrointestinal motility of mice with spleen deficiency and restore the gastrointestinal motility and lasting effect.Its mechanism of action is related to activation of Keap1/Nrf2/HO-1 signaling pathway and exerting anti-oxidative stress damage.
