Repairing Effects Of Residue Polysaccharides From Oudemansiella Radicata On CCl4-induced Chronic Liver Injury

The rational and effective use of mushroom residues can not only solve the problem of waste of resources and environmental pollution,but also promote the improvement of the edible fungus industry.In this experiment,the conditions of Oudemansiella radicata residue polysaccharides?RPS?were optimized,its monosaccharide composition and bond type were analyzed,and the anti-oxidation,anti-inflammatory,anti-fibrotic and anti-apoptotic effects of RPS on CCl4-induced chronic liver injury?CLI?mice were investigated.The results are as follows:?1?When optimal extraction conditions of RPS were water addition 40 times,extraction times 2.08 and alcohol precipitation time 24 h,the maximum theoretical extraction rate of RPS was 3.41%.In view of the actual operating conditions,the extraction process conditions were adjusted to water addition 40 times,extraction times 2 and alcohol precipitation time 24h,the predicted theoretical yield was 3.40%,and the actual RPS yield was 3.38%.?2?High performance liquid chromatography?HPLC?results showed that galactose?Gal?,xylose?Xyl?and arabinose?Ara?were the main monosaccharides of RPS,with a molar ratio of 2.25:2.72:2.34.Fourier transform infrared spectroscopy?FT-IR?analysis demonstrated that RPS had a characteristic absorption peak of polysaccharide,which was a pyranose containing?-type glycosidic bond.?3?The high-dose RPS?400 mg/kg?had the best liver protection effects,and could significantly increase the superoxide dismutase?SOD?,catalase?CAT?and glutathione peroxidase?GSH-Px?activities,and significantly reduce cytochrome P450 2E1?CYP2E1?,malondialdehyde?MDA?and lipid peroxidation?LPO?levels,and effectively inhibit lipid peroxidation in liver.Tumor necrosis factor-??TNF-??,interleukin-6?IL-6?and interleukin-1??IL-1??levels of liver were decreased significantly.Western blot results showed that RPS could reduce the level of the pro-inflammatory factor p-p65?phosphorylated p65?and increase the level of the inflammation inhibitor I-?B??Inhibitor of nuclear factor kappa-B??through the NF-?B pathway to alleviate the inflammatory response.RPS might alleviate liver fibrosis by reducing the level of TGF-?1?Transforming growth factor-?1?.Quantitative real-time polymerase chain reaction?qRT-PCR?results suggested that RPS down-regulated the relative mRNA expression of Bax and up-regulated Bcl-2?B-cell lymphoma-2?relative mRNA expression to achieve anti-apoptotic effect.In addition,histopathological observations of mice liver including hematoxylin and eosin?H&E?staining,Masson's trichrome staining,and Hoechst 33258 apoptotic staining demonstrated that RPS could repair the CCl4-induced chronic liver damage.The above results indicated that RPS had anti-oxidant,anti-inflammatory,anti-fibrotic and anti-apoptotic activities,and effectively improved the CCl4-induced hepatocyte damage.
