ObjectiveThe expression of S100A8/A9,IL-6 in serum,vitreous fluid,and aqueous humor of patients with type 2 diabetic retinopathy(DR)were quantitatively detected,and analyzed the correlation between S100A8/A9 with DR,IL-6 and other systemic factors,to explore whether S100A8/A9 is associated with DR pathogenesis,and provide new ideas and basis for the diagnosis and treatment of DR.Methods47 patients(47 eyes)who underwent cataract extraction and vitrectomy in our department from January 2019 to December 2019 were selected,among them 27 patients(27 eyes)with proliferative diabetic retinopathy(PDR)as experimental group,20 patients(20 eyes)with idiopathic macular hole(IMH)were used as control group,to collect their serum,vitreous and aqueous humor;25 patients(25 eyes)with non-proliferative diabetic retinopathy(NPDR)were selected as the experimental group to collect serum from patients.According to the medical history survey,collect about 5 ml of venous blood of all enrolled patients,the serum was separated and stored at-80? for testing.Vitreous was collected from patients with PDR and IMH about 0.2-0.4ml,after rapid centrifugation,collected supernatant and stored at-80? for measurement.Aqueous humor was collected from patients with PDR and IMH,and 0.1 mL of aqueous humor was extracted through anterior chamber puncture before surgery,and stored at-80? for testing.General data of all subjects were collected,such as gender,age,body mass index(BMI),duration of DM,age of onset of DM,duration of DR,and clinical indicators:fasting blood glucose(FPG),glycated hemoglobin(HbA1c),serum urea(SU),serum creatinine(SCr),triglyceride(TG),total cholesterol(TC),low density lipoprotein(LDL),high density lipoprotein(HDL)were measured.Using the enzyme-linked immunosorbent assay(ELISA)to quantitatively detect the concentrations of S100A8/A9,IL-6 in serum,aqueous humor and vitreous fluid samples,and the above data were collated and processed for statistical analysis.Results1.Comparison of general information and clinical indicators between the experimental group and the control group:There was no significant difference in gender,age,and body mass index between the NPDR group,the PDR group(experimental group),and the control group(P>0.05);The differences between groups in HbAlc,FPG,SU,TC,TG,LDL,HDL were statistically significant(P<0.05);The differences between NPDR group and PDR group in the course of DM,the course of DR,and the age of onset of DM were statistically significant(P<0.05).2.Comparison of S100A8/A9 concentration in serum,vitreous and aqueous humor between experimental group and control group:The concentration of S100A8/A9 in the serum of NPDR group,PDR group(experimental group)and control group was statistically significant(P<0.05),and the differences between any two groups in these three groups were statistical significance(P<0.05);The concentration of S100A8/A9 in vitreous of PDR group was significantly higher than that of control group,with statistical significance(t=4.523,P=0.000);The concentration of S100A8/A9 in aqueous humor of the PDR group was significantly higher than that of control group,and the difference was statistically significant(t=3.269,P=0.003);3.Comparison of S100A8/A9 concentration in serum,vitreous,aqueous humor of the experimental group and correlation analysis:The difference in S100A8/A9 concentration between the three groups was statistically significant(F=65.758,P=0.000);but there was no significant difference in the concentration of S100A8/A9 between vitreous and aqueous humor(P>0.05);The three are positively correlation with each other,serum and vitreous(r=0.741),serum and aqueous humor(r=0.642),vitreous and aqueous humor(r=0.501),and all were statistically significant(P<0.05);4.Correlation of S100A8/A9 concentration in serum,vitreous and aqueous humor with general information and clinical indicators in the experimental group:The serum concentration of S100A8/A9 of DR patients was positively correlated with HbA1c and DR course(r=0.460,r=0.390),and negatively correlated with HDL(r=-0.378);The concentration of S100A8/A9 in vitreous of PDR patients was positively c.orrelated with HbAlc,DR course,SU,and SCr(r=0.448,0.412,0.773 and 0.349),and negatively correlated with HDL(r=-0.327);The concentration of S100A8/A9 in aqueous humor of PDR patients was positively correlated with HbA1c,SU,SCr,and TC(r=0.396,0.315,0.350 and 0.349);5.Comparison of IL-6 concentration in serum,vitreous,aqueous humor between PDR group(experimental group)and control group and correlation between S100A8/A9 and IL-6:The concentration of IL-6 in serum,vitreous and aqueous humor of PDR group was significantly higher than that of control group,the difference was statistically significant(P<0.05);Serum,vitreous and aqueous humor S100A8/A9 were positively correlated with IL-6(r=0.452,0.738,0.651);Conclusion1.In the early stage of DR,S100A8/A9 level have begun to rise during systemic circulation.In the PDR stage,the levels of S100A8/A9 are significantly increased throughout the body and locally,suggesting that S100A8/A9 level reflect the severity of DR.It is preliminary indicated that S100A8/A9 may participate in the occurrence and development mechanism of DR,and may become a predictor of DR onset;2.The expression of S100A8/A9 is related to the course of DR,blood glucose,blood lipids,and renal function,S100A8/A9 may promote IL-6 secretion and participate in the formation of DR.
